干扰素诱导跨膜蛋白3基因rs12252多态性与新型冠状病毒肺炎关联性研究 被引量:3

摘要 目的:了解干扰素诱导跨膜蛋白3(IFITM3)基因rs12252多态性与新型冠状病毒感染之间的关联。方法:选择2020年1月至2021年1月北京市监测发现的新型冠状病毒肺炎病例及无症状感染者446例和健康对照人群65例。提取呼吸道样本基因组DNA,使用Sanger测序法检测IFITM3基因rs12252多态性。结果:无症状感染者、轻型病例、普通型病例中rs12252基因型分布频率与对照组无显著性差异(P值均>0.05),但重型病例中基因型分布具有显著差异(P<0.05)。60%的重型病例为CC基因型,明显高于其他类型病例、无症状感染者或健康人群。隐性遗传模型显示CC基因型个体比TT基因型和CT基因型个体有更高的风险发展为新冠肺炎重型或危重型(OR=3.546,95%CI:1.084~11.595)。结论:IFITM3 rs12252CC基因型会导致较高的新冠肺炎重症感染风险。 Objective To investigate the relationship between rsl2252 polymorphism of interferon-induced transmembrane protein 3(IFITM3)gene and severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)infection.Methods A total of 446 new cases of coronavirus disease 2019(COVID-19)and asymptomatic carriers identifled by the surveillance in Beijing from January 2020 to January 2021,and 65 healthy control individuals were enrolled in this study.The genomic DNA was extracted from respiratory samples.The polymorphism of rsl2252 in IFITM3 gene was determined by Sanger sequencing.Results No statistically significant differences in distribution of rs 12252 polymorphism in IFITM3 was observed between controls and asymptomatic carriers,mild cases,or common cases(P>0.05).However,the difference between controls and severe cases was statistically significant(P<0.05).The CC-genotype was observed in 60%of severe cases,which was significant higher than that of other types of cases,asymptomatic carriers,and controls.Recessive genetic model suggested that COVID-19 cases with CC genotype had higher risk of developing into severe or critical cases than those with CT or TT genotype(072=3.546,95%CI:1.084-11.595).Conclusions The CC genotype of rs 12252 in IFITM3 gene may related to higher risk of developing into severe or critical cases of COVID-19.
