摘要
目的:采用蛋白组学技术研究口腔溃疡及其阴虚火旺、胃火炽盛证的血浆代谢特征。方法:经iTRAQ-2D和LC-MS/MS分析,结合生物信息学分析获得差异蛋白质,利用脂质组学技术分析血浆脂质含量变化。结果:经蛋白组学分析,鉴定出口腔溃疡患者体内61种差异蛋白质,其中炎症因子C-反应蛋白(CRP)、血清沉淀样蛋白A(SAA)、载脂蛋白APOC2、APOM、APOL1和磷脂酰肌醇(PI)等上调,热休克蛋白HSP90、14-3-3和磷脂酰乙醇胺(PE)等下调,凝血因子Ⅻ(F12)在阴虚火旺型患者体内显著升高(P<0.05),补体C1q在胃火炽盛型患者体内显著升高(P<0.05)。结论:口腔溃疡患者体内凝血补体级联系统、糖代谢和脂质代谢等发生特异性变化,且阴虚火旺、胃火炽盛型患者在这些代谢途径中具有不同的代谢特征。
Objective: To study the plasma metabolic characteristics of oral ulcer and its yin deficiency with effulgent fire and intense stomach fire syndrome by proteomics. Methods: By iTRAQ-2 D and LC-MS/MS analysis, combined with bioinformatics analysis, the differential proteins were obtained. Results: By proteomic analysis, 61 differential proteins were identified in patients with oral ulcer, including inflammatory factor C-reactive protein(CRP), serum precipitation like protein A(SAA), apolipoprotein apoc2, Apo M, APOL1 and phosphatidylinositol(PI) up-regulated, heat shock protein HSP90, 14-3-3 and phosphatidylethanolamine(PE) down regulated, the level of coagulation factor Ⅻ(F12) was significantly increased in patients with yin deficiency with effulgent fire(P<0.05). and complement C1q was significantly increased in patients with intense stomach fire syndrom(P<0.05). Conclusion: There are specific changes in coagulation complement cascade system, glucose metabolism and lipid metabolism in patients with oral ulcer, and patients with yin deficiency with effulgent fire and intense stomach fire have different metabolic characteristics in these metabolic pathways.
