摘要
探究马铃薯内生真菌Aspergillus carneus提取物中丁烯内酯类化合物Asperteretal D对乳腺癌细胞(MCF-7)的凋亡诱导作用及其分子机制.结果表明:化合物Asperteretal D剂量依赖性地促进MCF-7细胞凋亡,分子对接实验表明其能够与PARP、Caspase-9、P53、Bcl-2、Bax等凋亡相关蛋白结合.化合物Asperteretal D能显著下调pro Caspase-9、pro Caspase-3、PARP蛋白的表达,同时上调P53蛋白的表达.因此,化合物Asperteretal D通过诱导乳腺癌细胞凋亡,从而显著抑制其增殖.
To explore the tumor cell apoptosis-inducing effect and mechanism of butene lactone compound Asperteretal D,extracting from potato endophytic fungus Aspergillus carneus.Asperteretal D promoted apoptosis of MCF-7 cell in a dose-dependent manner.The molecular docking results showed that Asperteretal D can bind to PARP,Caspase-9,P53,Bcl-2,Bax and other apoptosis-related proteins.Asperteretal D could significantly down-regulate the expression of pro Caspase-9,pro Caspase-3 and PARP proteins,meanwhile up-regulate the expression of P53 protein.Compound Asperteretal D can induce apoptosis of MCF-7 cells,and its molecular mechanism is closely related to caspase cascade and tumor suppressor P53 protein.Compound Asperteretal D can significantly inhibit the proliferation of breast cancer cells by inducing apoptosis.
