桦褐孔菌醇提物对大鼠心肌缺血再灌注损伤的保护作用

On the protective effect of the inonotus obliquus alcohol extract on the myocardial ischemia-reperfusion injury in rats

摘要 目的观察桦褐孔菌醇提物对大鼠心肌缺血再灌注损伤的影响,并初步探究其潜在的保护作用机制。方法健康雌性Wistar大鼠125只,随机分为假手术组、模型组和桦褐孔菌醇提物150、300、600 mg·kg^(-1)治疗组,每组25只。假手术组进行手术操作但不结扎,其他组大鼠进行结扎缺血处理2 h,再灌注2 h。再灌注2 h后,采用彩色多普勒超声便携机检测大鼠心脏功能,测定大鼠心肌梗死面积,检测血清中CK-MB、AST和LDH活性,分光光度法检测心肌组织中SOD、CAT、GSH-px活性和MDA含量。结果桦褐孔菌醇提物能明显改善大鼠心脏功能,减少心肌梗死面积,降低大鼠血清中CK-MB、AST和LDH活性,升高心肌组织SOD、CAT和GSH-px活性,降低MDA含量。结论桦褐孔菌醇提物能够提高心肌抗氧化应激,对大鼠心肌缺血再灌注损伤起到保护作用。 Objective To observe the effect of the inonotus obliquus alcohol extract on the myocardial ischemia-reperfusion injury in rats,and to preliminarily explore its potential protective mechanism.Methods A total of 125 healthy female Wistar rats were randomly divided into sham operation group,model group,and inonotus obliquus alcohol extract 150,300,and 600 mg·kg^(-1) treatment groups,with 25 rats in each group.The rats in the sham operation group were operated without ligation.The rats in the other groups were treated with ligation and ischemia for 2 hours,and with reperfusion for 2 hours.After the reperfusion,the cardiac function of the rats was detected by the color Doppler ultrasound portable machine,the area of myocardial infarction was measured,the activity of CK-MB,AST and LDH in serum was detected,and the activity of SOD,CAT,GSH-px and the MDA content were detected by spectrophotometry.Results The alcohol extract of inonotus obliquus significantly improved the cardiac function of rats,reduced the area of myocardial infarction,reduced the activity of CK-MB,AST and LDH in the rat serum,increased the activity of SOD,CAT and GSH-px of the myocardial tissue,and reduced the MDA content.Conclusion The alcohol extract of inonotus obliquus can improve the myocardial antioxidant stress and protect the myocardial ischemia-reperfusion injury in rats.
