8个新收集大白菜杂交种DNA指纹构建及其与已有种质的亲缘关系分析

摘要 为了不断充实大白菜杂交种DNA指纹数据库,了解新收集种质与已有种质的亲缘关系并加以快速利用,本研究利用前期开发的大白菜杂交种指纹构建核心引物,对新收集的8个大白菜杂交种进行PCR扩增,构建了8个新杂交种的DNA指纹,丰富了大白菜杂交种DNA指纹数据库。采用MEGA7.0软件对新品种和已有品种及育种材料的指纹数据进行比对分析,发现新收集品种与之前收集的品种或创制的材料虽然有相似之处,但并非完全相同。这表明新收集的品种与之前创制的材料或收集的品种有不同的遗传背景,可以作为新种质加以利用来拓宽现有种质的基因谱。同时也表明,DNA指纹技术可以用于对不同种质的比较研究。 In order to enrich the DNA fingerprint database of Chinese cabbage hybrids, and to learn about the genetic relationships among all collected germplasms, the core primers developed in our previous studies were used for PCR amplification of 8 new collected Chinese cabbage hybrids. DNA fingerprint of the 8 Chinese cabbage hybrids was constructed and the results expanded the DNA fingerprint database of Chinese cabbage hybrids. The data of DNA fingerprint of the 8 new collected Chinese cabbage hybrids and previously collected germplasms were compared by software MEGA 7.0. It revealed that there were some similarities between the 8 new hybrids and previously detected germplasms or hybrids, but there were no entirely homologous ones. The results showed that the new collections were different in genetic background from previous created germplasms or collected hybrids. They were useful for broadening the gene pool of germplasms now available. It also suggested that DNA fingerprint could be used in analyzing the genetic relationship of different germplasms.
