摘要
目的:探讨蓝萼香茶菜总二萜(total diterpenes of cycalyx herb,TD)对实验性兔心肌缺血/再灌注损伤相关蛋白的保护作用。方法:将健康日本大耳白兔18只分为3组,A:假手术(Sham)组;B:缺血/再灌注(I/R)组;C:TD组。光镜和电镜下观察3组兔心肌细胞结构变化情况;检测再灌注180 min时3组兔血清丙二醛(MDA)、超氧化物歧化酶(SOD)及心肌MDA、SOD、谷胱甘肽过氧化物酶(GSH-Px)和Caspase 3活性变化情况;WB法检测3组兔心肌细胞凋亡蛋白Bcl-2和Bax的含量;评价TD预处理对在体兔心肌缺血/再灌注损伤(MI/RI)动物模型的保护作用。结果:TD组与I/R组相比:血清MDA、心肌MDA和Caspase 3下降(P<0.05),血清SOD、心肌SOD和GSH-Px增加(P<0.05),Bax蛋白表达下降(P<0.05),Bcl-2蛋白表达增加(P<0.05)。结论:TD能减少实验性兔MI/RI的促凋亡蛋白Bax的表达,增加抗凋亡蛋白Bcl-2的表达,提示该药对心肌细胞凋亡有保护作用。
Objective:To survey the protective effects of total diterpenes(TD)of cycalyx herb on myocardial ischemia/reperfusion(I/R)related protein in rabbits.Methods:All 18 rabbits were separated into three groups:A:sham operation group(Sham);B:ischemia/reperfusion(I/R)group;and C:TD group.The changes in myocardial cells structure of the rabbits in three groups were observed under light microscope and electrical microscope;the changes in MDA and SOD activity in the serum,MDA,SOD,GSH-Px and Caspase 3 activity in myocardial cells were measured after reperfusion for 180 minutes;the contents of myocardial cells apoptosis-related protein Bcl-2 and Bax in rabbit myocardial cells were detected using WB method;the protective effects of TD preconditioning on MI/RI rabbit model were assessed.Results:When TD group was compared with I/R group:serum MDA,myocardial MDA and Caspase 3 reduced(P<0.05),serum SOD,myocardial SOD and GSH-Px increased(P<0.05),Bax protein expressions lowered(P<0.05),Bcl-2 protein expression increased(P<0.05).Conclusion:TD could reduce apoptosis-related protein Bax expression and increase anti-apoptosis protein Bcl-2 expression,and it is suggested that TD has the protective effects on the apoptosis of myocardial cells.
