白细胞介素-6在皮肤损伤修复中的作用及对炎症因子表达水平的影响研究 被引量:6

摘要 目的:探讨白细胞介素-6(IL-6)在皮肤损伤修复中的作用及对炎症因子表达水平的影响。方法:选择2017年5月-2017年12月参与实验的SD大鼠30只作为对象,随机取10只,设为常规对照组;剩余20只大鼠采用手术方式在脊柱两侧1cm部位作皮肤缺损损伤动物模型,建模完毕后随机将SD大鼠分为IL-6基因组(n=10)和模型对照组(n=10)。常规对照组与模型对照组常规饲养、饮水,IL-6基因组建模后腹腔注射IL-6基因转移的高分泌IL-6 0.5ml,注射1次。采用酶联免疫吸附试验、PCR方法完成三组大鼠白细胞介素-1α(IL-1α)、白细胞介素-1β(IL-1β)、角质细胞诱导因子(KC)、单核细胞诱导蛋白1α(MIP-1α)及单核细胞诱导蛋白-2(MIP-2)水平及基因测定,比较三组损伤面积、炎症因子水平、炎症因子基因表达水平。结果:IL-6基因组干预后1d、3d、6d、10d及14d损伤面积缩小率大于模型对照组,差异有统计学意义(P<0.05)。IL-6基因组干预后IL-1α、IL-1β、KC、MIP-1α及MIP-2水平,低于模型对照组但高于常规对照组,差异有统计学意义(P<0.05)。模型对照组干预后IL-1α、IL-1β、KC、MIP-1α及MIP-2水平高于常规对照组,差异有统计学意义(P<0.05)。IL-6基因组IL-1α、IL-1β、KC、MIP-1α及MIP-2 基因表达量,均低于模型对照组,差异有统计学意义(P<0.05)。结论:IL-6在大鼠皮肤缺损中能降低测定炎症因子的产生,有助于促进皮肤损伤的修复,缩小损伤面积,促进大鼠恢复。 Objective To investigate the effect of interleukin-6(IL-6)on the expression of inflammatory factors in skin injury repair.Methods Thirty SD rats from May 2017 to December 2017 were selected as the subjects,and 10 rats were randomly selected as the conventional control group.The remaining 20 rats were surgically performed on the 1cm side of the spine.Animal models of skin defect injury were randomly divided into the IL-6 gene group(n=10)and the model control group(n=10)after modeling.The conventional control group and the model control group were routinely fed and drinking water.After the IL-6 gene group modeling,0.5ml of high-secretion IL-6 gene of IL-6 gene transfer was intraperitoneally injected and injected once.Three groups of rats were treated with interleukin-1α(IL-1α),interleukin-1β(IL-1β),keratinocyte inducing factor(KC),and monocyte-inducible protein 1a by enzyme-linked immunosorbent assay(ELISA)and PCR.(MIP-1α)and monocyte-inducible protein-2(MIP-2)levels and gene assays,comparing the three groups of lesion area,inflammatory factor levels,inflammatory factor gene expression levels.Results The reduction rate of injury area in the IL-6 gene group were higher than those in model control group on 1d,3d,6d,10d and 14d,and the differences were statistically significant(P<0.05).The levels of IL-1α,IL-1β,KC,MIP-1αand MIP-2 after the IL-6 gene group were lower than those in the model control group,but higher than those in the conventional control group(P<0.05).The levels of IL-1α,IL-1β,KC,MIP-1αand MIP-2 in the model control group after intervention were higher than those in the conventional control group(P<0.05).The levels of IL-1α,IL-1β,KC,MIP-1αand MIP-2 were higher in the model control group than in the conventional control group(P<0.05).The gene expression levels of IL-1α,IL-1β,KC,MIP-1αand MIP-2 in the IL-6 gene group were lower than those in the model control group(P<0.05).Conclusion IL-6 can reduce the production of inflammatory factors in rat skin defects,help to promote the repair of skin damage,reduce the damage area and promote the recovery of rats.
