补肾活血祛痰方对高雄激素多囊卵巢综合征大鼠类固醇激素合成急性调节蛋白质因子的影响

Effect of Bushen Huoxue Qutan Recipe on Star protein factor in hyperandrogen induced PCOS rats

目的:探讨补肾活血祛痰方对高雄激素多囊卵巢综合征(PCOS)大鼠类固醇激素合成急性调节蛋白(StAR)的影响。方法:选择SD雌性大鼠60只,采用脱氢表雄酮注射建立高雄激素PCOS模型,并随机分为对照组(30只)和中药组(30只),中药组给予补肾活血祛痰方灌胃治疗,对照组给予等量的0.9%氯化钠溶液灌胃,观察治疗后两组大鼠卵巢组织形态学情况,比较治疗后两组大鼠血清激素水平情况;Western blot法检测治疗后两组大鼠StAR蛋白的表达情况。结果:治疗后,卵巢组织形态学观察结果显示,对照组仍存在多个囊泡,其囊泡即空卵泡,中药组卵巢组织形态明显改善;与对照组相比,卵泡及黄体数较多(均P<0.05);治疗后,中药组血清促黄体生成素(LH)和雄激素(T)水平比对照组明显降低(均P<0.05)。治疗22 d后,中药组血清胰岛素样生长因子-Ⅰ(IGF-Ⅰ)和肿瘤坏死因子(TNF-α)水平比对照组明显降低(均P<0.05)。Western blot结果显示,治疗后中药组StAR蛋白表达明显比对照组降低(P<0.05)。结论:补肾活血祛痰方可改善高雄激素PCOS大鼠卵巢功能,促进激素水平趋于正常,并可降低卵泡膜细胞的细胞膜附近StAR蛋白质,改善大鼠的生殖功能。

Objective:To investigate the effect of Bushen Huoxue Qutan recipe on acute regulatory protein(StAR)of steroid hormone synthesis in rats with polycystic ovary syndrome(PCOS).Methods:60 female SD rats were randomly divided into control group(n=30)and traditional Chinese medicine group(n=30).The traditional Chinese medicine group was given Bushen Huoxue Qutan decoction by gavage,while the control group was given the same amount of normal saline by gavage Western blot was used to detect the expression of star protein.Results:After treatment,the observation of ovarian histomorphology showed that there were still multiple follicles in the control group,which were empty follicles.The ovarian histomorphology in the traditional Chinese medicine group was significantly improved;compared with the control group,there were more follicles and corpus luteum at all levels(all P<0.05);after 22 days of administration,the serum levels of LH and T in the TCM group were significantly lower than those in the control group(all P<0.05).After 22 days of treatment,serum IGF-Ⅰand TNF-αlevels in the TCM group were significantly higher than those in the control group compared with the control group,the serum creatinine level was significantly lower(all P<0.05).Western blot results showed that the expression of star protein in the traditional Chinese medicine group was significantly lower than that in the control group 22 days after administration(P<0.05).Conclusion:Bushen Huoxue Qutan decoction can improve the ovarian function of PCOS rats with hyperandrogen,promote the hormone level to normal,and reduce the StAR protein near the cell membrane of follicular membrane cells.
