人参皂苷对糖尿病肾病大鼠体内ColⅣ、Nrf2和NQO1表达的影响 被引量:4

摘要 目的研究人参皂苷对糖尿病肾病大鼠体内Ⅳ型胶原蛋白(ColⅣ)、Nrf2和醌氧化还原酶1(NQO1)表达的影响。方法30只雄性SD大鼠随机分为正常对照组(NC组),糖尿病肾病模型组(DN组),糖尿病肾病模型加人参皂苷治疗组(MG组),每组10只。对3组大鼠的体重、血糖和生化指标分别进行记录;HE染色观察大鼠肾组织形态;Western blot检测大鼠ColⅣ、Nrf2和NQO1蛋白并比较,用RT-PCR法检测mRNA的相对表达量。结果实验前3组大鼠体重差异无统计学意义(P>0.05),实验后与NC组大鼠的体重和血糖比较,DN组大鼠显著降低,血糖值显著升高(P<0.05);与DN组大鼠比较,GM组大鼠的体重和血糖值明显得到改善(P<0.05)。DN组生化指标的浓度明显高于NC组,而Ins明显降低(P<0.05);和DN组生化指标比较,GM组的浓度明显降低,Ins明显升高(P<0.05);与NC组大鼠的肾组织形态比较显示,DN组大鼠的情况明显较差,并且肾小球系膜细胞与高度增生出现;GM组大鼠的肾组织形态比DN组大鼠有明显的改善。蛋白和mRNA比较显示,DN组大鼠ColⅣmRNA和蛋白的表达均比NC组显著增高(P<0.05),Nrf2、NQO1表达明显减少,MG组和DN组大鼠比较,经过人参皂苷治疗后,其ColⅣ表达量明显降低,Nrf2、NQO1表达量显著升高(P<0.05)。结论人参皂苷可以抑制大鼠体内ColⅣ的表达,提高Nrf2和NQO1蛋白的表达,从而改善糖尿病肾病大鼠血糖及肾功能水平。 Objective To investigate the effects of Ginsenoside on the expression of ColⅣ,Nrf2 and NQO1 in rats with diabetic nephropathy.Methods Thirty male SD rats were randomly divided into control group(NC group),diabetic nephropathy model group(DN group)and diabetic nephropathy model+ginsenoside treatment group(MG group),with 10 rats in each group.The weight,blood glucose and biochemical indexes were observed,and HE staining was used to observe the morphology of the rat kidney.Moreover,Western Blot was used to detect the levels of ColⅣ,Nrf2 and NQO1 protein.In addition,RT PCR was used to determine the relative expression levels of mRNA.Results Before the experiment,there was no significant difference in the weight of rats among the three groups(P>0.05).After experiment,the body weight in DN group was significantly lower than that in NC group,however,the blood glucose levels were significantly higher than those in NC group(P<0.05).The weight and blood glucose in GM group,compared with those in DN group,were significantly improved(P<0.05).Moreover,the levels of biochemical indexes in DN group were significantly higher than those in NC group,while Ins levels were significantly reduced(P<0.05).The levels of biochemical indexes in GM group,compared with those in DN group,were significantly decreased,while Ins levels were significantly increased(P<0.05).In addition,the nephridial tissue morphous in DN group was inferior to that in NC group,however,which in GM group was significantly improved.In addition the expression levels of ColⅣmRNA and protein in DN group were significantly higher than those in NC group(P<0.05),but the expression levels of Nrf2 and NQO1 were significantly decreased.After treatment with Ginsenoside,the expression levels of ColⅣwere significantly decreased,however,the expression levels of Nrf2 and NQO1 were significantly increased(P<0.05).Conclusion Ginsenoside can improve the blood sugar and renal function of rats with diabetic nephropathy by inhibiting the expression of ColⅣand increasing the expressions of Nrf2 and NQO1 protein.
