Effects of Roux-en-Y gastric bypass on behavior patterns and energy expenditure in diabetic rats

摘要 目的:探讨RYGB对2型糖尿病大鼠行为模式、生理应激、能量代谢的影响。方法:将2型糖尿病的GK大鼠模型随机分为两组,RYGB组(行RYGB,n=5)与Sham组(行假手术,n=5),另外将年龄匹配的正常Wistar大鼠作为正常对照组(对照组,n=5)。分别于术前及术后2周、4周测量体重、空腹血糖。术后3周应用大鼠代谢监测系统分别对每只动物日间与夜间的行走活动、精细运动、水摄入量、食物摄入量、氧消耗及产热进行监测与分析。结果:与Sham组相比,RYGB组动物体重减轻,夜间精细运动、行走活动量、水摄入量及食物摄入量减少;RYGB组在日间与夜间的氧消耗量、产热均增加。结论:RYGB术后个体主动活动量与进食摄入减少,但代谢率与能量消耗增加,提示RYGB术后运动指导对进一步改善临床结局的重要性。 Objective:To investigate the effects of Roux-en-Y gastric bypass(RYGB)on physical activity pattern,physiological stress level,and subsequent energy consumption.Methods:Goto-Kakizaki(GK)type 2 diabetes rats were employed and randomized into two groups according to the surgical procedure performed including RYGB(n=5)and Sham surgery(n=5).In addition,age-matched Wistar rats were adopted as normal non-diabetic controls(n=5).Body weight and fasting plasma glucose were measured in vivo before surgery,2 weeks after surgery and 4 weeks after surgery.Day and night behavior parameters including ambulatory movement,fine movement,water consumption,food consumption,oxygen consumption and heat production were also monitored and analyzed in all animals in the 3rd week postoperatively.Results:RYGB significantly lowered the body weight at all the time points postoperatively comparing with sham animals.With regard to behavior parameters,it was found that RYGB,but not sham,had less amounts of fine movement,ambulatory movement,water consumption and food consumption in the night time postoperatively.However,oxygen consumption and heat production increased in RYGB group compared with sham animals during both daytime and night,indicating that RYGB animals experienced elevated energy expenditure and elevated metabolic rate even with insufficient energy intake and exercise.Conclusions:Physical activities and food intake are attenuated after RYGB procedure,while energy consumption is increased in GK rats.These findings imply that physical exercise coach should be a favorable supplement after RYGB that may guarantee improved outcome.
