摘要
目的探讨干扰素诱导跨膜蛋白2(IFITM2)在胃癌组织中的表达及其对胃癌细胞迁移和侵袭的影响。方法收集南方医科大学顺德医院2012年1月至2015年1月收治的54例胃癌患者癌组织及对应的癌旁组织样本,检测胃癌与癌旁组织、胃癌细胞MKN45与正常胃黏膜上皮细胞GES-1中IFITM2的表达水平,分析胃癌组织中IFITM2的表达与临床病理特征及预后的关系。沉默IFITM2表达,检测MKN45细胞的迁移和侵袭能力,及对E-cadherin和Vimentin表达水平的影响。结果胃癌组织及MKN45细胞中IFITM2的表达水平明显高于癌旁组织(P<0.01),胃癌组织中IFITM2的表达水平与TNM分期、肿瘤浸润程度、淋巴结及远处转移情况相关(P<0.05),并与患者的预后不良相关(P<0.05)。沉默IFITM2后,MKN45细胞的迁移和侵袭能力降低(P<0.05),并能上调E-cadherin的表达和抑制Vimentin的表达(P<0.05)。结论IFITM2在胃癌组织和细胞中高表达,通过调控E-cadherin和Vimentin的表达而促进胃癌细胞的迁移和侵袭,并与患者的预后不良显著相关。
Objective To investigate the IFITM2 expression in gastric cancer tissues and its effect on gastric cancer cell migration and invasion.Methods From January 2012 to January 2015,54 patients with gastric cancer were collected from Shunde Hospital of Southern Medical University.The expression level of interferon-induced transmembrane 2(IFITM2)in gastric cancer and adjacent tissues,gastric cancer cell MKN45 and normal gastric mucosa epithelial cell GES-1 was detected,and the relationship between IFITM2 expression in gastric cancer and clinicopathological features and prognosis was analyzed.The expression of IFITM2 was silenced,and the migration and invasion ability of MKN45 cells were detected,as well as the effects on the expression levels of E-cadherin and Vimentin.Results The expression level of IFITM2 in gastric cancer and MKN45 cells was significantly higher than that in adjacent tissues(P<0.01).The expression level of IFITM2 in gastric cancer was correlated with TNM stage,tumor invasion degree,lymph node and distant metastasis(P<0.05),and also with poor prognosis(P<0.05).The migration and invasion ability of MKN45 cells decreased(P<0.05),and the expression of E-cadherin was up-regulated and the expression of Vimentin was inhibited(P<0.05).Conclusion IFITM2 is highly expressed in gastric cancer tissues and cells,and promotes migration and invasion of gastric cancer cells by regulating the expression of E-cadherin and Vimentin,which is significantly related to poor prognosis of patients.
