干酪乳杆菌YQ336高密度培养条件优化

Optimization of high density culture conditions of L.casei YQ336

研究对L. casei YQ336进行高密度培养,通过单因素和响应面试验优化L. casei YQ336培养基和培养条件,绘制48 h内生长曲线。结果显示,L.casei YQ336最佳培养基组成为红糖20 g、酵母浸粉5 g、磷酸氢二钾5 g、绿豆∶黄豆∶甘薯为2∶1∶1的增殖液1 000 mL。高密度培养L. casei YQ336的最优培养条件为接种量7%、培养温度35℃、初始pH值5.5、培养时间20 h。在该发酵条件下,L. casei YQ336发酵后活菌数达1.58×1010CFU/mL。

The experiment conducted high-density culture of L.casei YQ336,optimized the medium and culture conditions of L.casei YQ336 through single factor and response surface experiments,and plotted the growth curve within48 h.The results showed that the optimal medium composition was brown sugar 20 g,yeast extract 5 g,K2HPO45 g,mung bean∶soybean∶sweet potato 2∶1∶1 proliferating solution 1 000 m L.The optimal culture conditions were inoculation amount 7%,culture temperature 35℃,initial p H value 5.5,and culture time 20 h.The viable number of L.casei YQ336after fermentation was 1.58×1010CFU/m L.
