摘要 通过网络药理学的方法探讨知母治疗老年痴呆症的作用机制,通过TCMSP数据库筛选知母的有效活性成分及作用靶点、Uniprot数据库查询作用靶点的基因名称、Drugbank数据库查询老年痴呆症的疾病靶点、Cytoscape3.6.1软件构建"药物-有效成分-靶点-疾病"网络图;通过String数据库构建药物-疾病靶点PPI网络;最后通过Metascape数据库进行基因本体GO和KEGG通路富集分析.从知母中筛选出有效活性成分15个,涉及相关靶点111个;筛选疾病靶点46个,二者共有靶点14个,包括CHRM1、CHRM2、CYP3A4、CHRM3、OPRD1等.GO分析得到分子功能包括G蛋白偶联胺受体活性、肾上腺素能受体活性等;细胞组成包括突触前膜的固有成分、突触后膜的组成部分等;生物过程包括腺苷酸环化酶激活肾上腺素能受体信号通路、肾上腺素能受体信号通路等;KEGG富集分析主要包括钙信号通路、cGMP-PKG信号通路、cAMP信号通路等.通过分析认为,知母治疗老年痴呆症可能是通过多成分、多靶点、多通路的方式来发挥作用. In this paper, the mechanism of anemarrhena asphodeloides in the treatment of Alzheimer’s disease based on network pharmacology method was explored.TCMSP database was used to screen the active ingredients and targets of Anemarrhena asphodeloides.Uniprot database was used to query the gene names of the targets.Drugbank database was used to query the disease targets of Alzheimer’s disease.The network diagram of "drug-active ingredients-target-disease" was constructed by Cytoscape3.6.1 software.PPI network of drug disease targets was constructed by using String database.Finally, enrichment analysis of Gene Ontology(GO) and KEGG pathway were carried out by using metascape database.15 active components were screened from Anemarrhena asphodeloides, involving 111 related targets.46 disease targets were screened. They have 14 targets, including CHRM1,CHRM2,CYP3A4,chrm3,OPRD1,etc.Through GO analysis, molecular functions including G protein coupled amine receptor activity and adrenergic receptor activity were obtained.Cell composition includes the intrinsic components of presynaptic membrane and the components of postsynaptic membrane.Biological processes include adenylate cyclase activated adrenergic receptor signaling pathway, adrenergic receptor signaling pathway and so on.KEGG enrichment analysis mainly includes calcium signaling pathway, cGMP PKG signaling pathway and cAMP signaling pathway and so on.Through the analysis, we believe that Anemarrhena asphodeloides may play a role in the treatment of Alzheimer’s disease through multi-component, multi-target and multi-channel way.
