金欣口服液对呼吸道合胞病毒诱发支气管哮喘模型小鼠血浆短链脂肪酸的影响 被引量:3

Effects of Jinxin Koufuye(金欣口服液)on plasma short-chain fatty acids in respiratory syncytial virus-induced bronchial asthma in model mice

摘要 目的研究金欣口服液对呼吸道合胞病毒(Respiratory Syncytial Virus,RSV)诱发支气管哮喘(以下简称哮喘)模型小鼠血浆短链脂肪酸(Short Chain Fatty Acids,SCFA)的影响。方法将40只SPF级BALB/c小鼠按随机数字表法分为正常组、模型组、泼尼松组、金欣口服液组。使用卵清蛋白(OVA)和氢氧化铝(alum)致敏、RSV诱发建立哮喘小鼠模型,正常组和模型组给予生理盐水,泼尼松组、金欣口服液组分别给予泼尼松和金欣口服液灌胃治疗。通过气相色谱-质谱仪的方法测定各组血浆SCFA含量,并观察金欣口服液、泼尼松干预对血浆SCFA水平的影响。结果待测SCFA混合标准品溶液中各组分和质控样本的相对标准偏差均小于30%,各待测SCFA在线性范围内的线性良好,相关系数均>0.99,提示数据具有良好的稳定性及重复性。与正常组小鼠相比,模型组血浆中部分SCFA水平明显降低,差异均有统计学意义,分别为乙酸(P<0.01)、丁酸(P<0.01)、SCFA总量(P<0.05)。而经过药物干预后,泼尼松组、金欣口服液组小鼠血浆中丁酸、SCFA总量较模型组升高,其中丁酸泼尼松组与金欣口服液组均P<0.01;SCFA总量泼尼松组P<0.05,金欣组P<0.01,金欣口服液组小鼠血浆中乙酸较模型组升高(P<0.01)。另外,泼尼松组小鼠血浆中戊酸较模型组降低(P<0.05)。其他SCFA含量在各组间比较中,差异无统计学意义(P>0.05)。结论金欣口服液可上调RSV诱发哮喘模型小鼠血浆中乙酸、丁酸及SCFA总含量至正常水平,表明金欣口服液缓解RSV诱发哮喘的部分作用机制可能与调节血浆中SCFA含量有关。 Objective To study the effects of Jinxin Koufuye(金欣口服液)on plasma short-chain fatty acids(SCFA)in respiratory syncytial virus(RSV)-induced bronchial asthma(hereinafter referred to as asthma)in model mice.Methods Forty SPF grade BALB/c mice were divided into normal group,model group,prednisone group and Jinxin Koufuye group.The establishment of asthma model mice were sensitized with a mixture of ovalbumin and aluminum hydroxide and induced by RSV.Normal group and model group were given normal saline,prednisone group and Jinxin Koufuye group were given prednisone and Jinxin Koufuye by intragastric administration,respectively.The content of plasma SCFA was determined by gas chromatography-mass spectrometry(GC-MS),and the effects of Jinxin Koufuye and prednisone on the level of plasma SCFA were observed.Results The relative standard deviation of each component and quality control sample in the mixed standard solution of SCFA to be tested was less than 30%.The linear range of SCFA to be tested was good,and the correlation coefficient was more than 0.99,indicating that the data had good stability and repeatability.Compared with the normal group,the partial levels of plasma SCFA in the model group significantly decreased,and the differences were statistically significant:acetic acid(P<0.01),butyric acid(P<0.01)and total amount of SCFA(P<0.05),respectively.After medicine intervention,the total amount of butyric acid and SCFA in plasma of prednisone group and Jinxin Koufuye group increased higher than those in model group,among them butyric acid in prednisone group and Jinxin Koufuye group was P<0.01 in both groups;The total amount of SCFA in prednisone group was P<0.05,and that in Jinxin koufuye group was P<0.01,the level of acetic acid in plasma in Jinxin Koufuye group increased higher than that in model group(P<0.01).In addition,the content of valeric acid in plasma of prednisone group decreased than that of model group(P<0.05).There was no significant difference in other SCFA content among the groups(P>0.05).Conclusion Jinxin Koufuye can up regulate the total content of acetic acid,butyric acid and SCFA in plasma of RSV-induced asthma in model mice to normal level,which indicates that part of the mechanism of Jinxin Koufuye in alleviating RSV-induced asthma may be related to regulating the content of SCFA in plasma.
