猪细小病毒病灭活疫苗佐剂筛选及配苗乳化工艺研究 被引量:2

摘要 为了筛选制备猪细小病毒病灭活疫苗(L株,悬浮培养)的佐剂,用注射用白油和法国赛比克公司生产的MontanideTMISA 206 VG佐剂(简称206佐剂)分别配制灭活疫苗,通过对两种疫苗的配苗乳化工艺、疫苗性状、无菌检验、安全检验、效力检验和免疫期等方面进行比较。结果显示:与注射用白油配制的疫苗相比,使用206佐剂配苗乳化工艺相对简单;制备疫苗剂型为水包油包水型,易注射,吸收良好;局部和全身均无不良反应,安全性良好;免疫后第7天猪细小病毒206佐剂疫苗免疫猪产生HI抗体效价显著高于白油佐剂疫苗,免疫后第60天猪细小病毒206佐剂疫苗免疫猪HI抗体效价达到峰值;免疫期为6个月。综上,确定206佐剂作为制备猪细小病毒病灭活疫苗的佐剂。 In order to screen and prepare the adjuvant for the preparation of inactivated porcine parvovirus vaccine(L strain, suspension culture), white oil for injection and MontanideTMISA 206 VG adjuvant(Here in after referred to as "206 adjuvant") produced by France Sebic Company were used respectively. The inactivated vaccine was prepared by comparing the emulsification process, vaccine characteristics, sterility test, safety test, efficacy test and immunization period of the two vaccines. The results showed that: compared with the vaccine prepared with white oil for injection, the emulsification process using adjuvant 206 was relatively simple;the preparation of the vaccine was a water-in-oil-in-water type, which was easy to inject and had good absorption;there were no local or systemic adverse reactions. Good safety;on the 7 th, after immunization, pigs immunized with porcine parvovirus 206 adjuvant vaccine produced significantly higher HI antibodies than that of the white oil adjuvant vaccine. On the60 th, after immunization, pigs immunized with porcine parvovirus 206 adjuvant vaccine produced a peak of HI antibodies;the immunity period was six months. In summary, it was determined that 206 adjuvant was used as the adjuvant for preparing inactivated porcine parvovirus vaccine.
