摘要 本文采用4种提取方法提取番木瓜中的草酸,并优化了提取方法;利用高效液相色谱建立了番木瓜中草酸的测定方法。结果表明,最佳提取条件为匀浆辅助提取,提取溶剂为0.2%磷酸水溶液,料液比为1∶7(g/mL),提取时间为2 min。番木瓜提取液经SAX固相萃取柱净化后,采用HPLC检测,草酸在0.5~50.0μg/mL的浓度范围内具有良好的线性关系,回收率为88.6%~90.5%,相对标准偏差为3.1%~5.8%;利用该方法对12个番木瓜样品中的草酸含量进行检测,得出草酸含量为17.2~69.0 mg/kg。因此,本文建立的HPLC检测方法操作简单、灵敏度高、重复性好,可用于番木瓜中草酸的测定。 In this paper,four extraction methods were used to extract oxalic acid in papaya and optimize the extraction method.The method was established for the determination of oxalic acid in papaya by HPLC.The optimized extraction solvent was 0.2%phosphoric acid solution,the extraction time was 2 min and the ratio of material liquid was 1∶7(g/mL).The extract was purified by SAX SPE and then separate by HPLC.The recoveries were ranged from 88.6%to 90.5%and the relative standard deviations were ranged from 3.1%to 5.8%.The 12 papaya samples were determined and the contents of oxalic acid was between 17.2 mg/kg and 69.0 mg/kg.The method was simple,highly sensitive and repeatable,which could be applied for the determination of oxalic acid in papaya.
