摘要
目的:通过对基因表达综合数据库(Gene expression omnibus,GEO)中甲状腺乳头状癌(Papillary thyroid carcinoma,PTC)样本的测序数据进行生物信息学分析,筛选出PTC新的潜在调控分子靶标,促进PTC的机制研究。方法:从GEO中搜索PTC及正常甲状腺组织样本的转录组测序数据,通过多种在线网站及R软件筛选出差异表达基因,进行功能富集分析及蛋白相互作用关系分析,寻找可能影响PTC发生发展的潜在核心基因。结果:共筛选出100个差异上调和60个差异下调基因,功能富集分析结果显示差异基因主要富集在蛋白质细胞外基质、钙离子结合、基底膜、细胞表面、胶原蛋白结合、肝素结合、兴奋性突触、细胞粘附等功能中。构建蛋白互作网络图提示FN1、SerpinA1、PLAU、APOE、TOP2A、CDH2、BUB1、ASPM、MKI67、APOC1等为互作网络的核心基因。经过肿瘤基因组图谱(The cancer genome atlas,TGCA)数据及人蛋白质数据分析,Top10核心基因中有3个为显著差异表达基因,即FN1、SerpinA1和CDH2。结论:甲状腺乳头状癌患者与健康人群的甲状腺组织的整体基因表达存在明显差异,差异基因FN1、SerpinA1、CDH2有望作为PTC机制研究、诊断的可靠分子标记物。
Objective:Through bioinformatics mining and analysis of the sequencing data of Papillary thyroid carcinoma(PTC)samples in the gene expression omnibus(GEO)database,new potential regulatory molecular targets of PTC are screened,thereby promoting the study of the mechanism of PTC.Methods:Search for the transcriptome sequencing data of PTC and control thyroid tissue samples from GEO,screen out differentially expressed genes through a variety of online websites and R software,perform functional enrichment analysis and protein interaction Analysis of the role relationship,looking for the potential core genes that may affect the occurrence and development of PTC.Results:A total of 100 differentially up-regulated and 60 differentially down-regulated genes in PTC were screened.The results of functional enrichment analysis showed that the differential genes were mainly enriched in protein extracellular matrix,calcium ion binding,basement membrane,cell surface,collagen binding,and heparin binding,excitatory synapse,cell adhesion and other functions.The construction of protein interaction network diagram suggested that FN1,SerpinA1,PLAU,APOE,TOP2A,CDH2,BUB1,ASPM,MKI67,APOC1,etc.were the core genes of the interaction network.After transcriptome sequencing data verification,three of the Top10 core genes were differentially expressed,namely FN1,SerpinA1 and CDH2.Conclusion:There are significant differences in the overall gene expression of thyroid tissue between PTC patients with papillary thyroid carcinoma and healthy people.The differential genes FN1,SerpinA1,and CDH2 are expected to serve as molecular markers for PTC mechanism research,diagnosis and prognosis.
