基于高通量测序的野百合转录组分析

Transcriptome Analysis for Lilium brownii Based on High Throughput Sequencing

摘要 本研究基于新一代高通量测序技术平台Illumina HiSeq^(TM) 4000对野百合进行转录组测序,对物种的转录组序列进行统计,并将得到的数据进行de novo组装,结果共获得47605条Unigenes,总长度为33972306 bp,平均长度为713 bp,N50为1204 bp。将获得的Unigenes与Nr、Swiss-Prot、KEGG以及KOG数据库进行比对,结果显示,分别有28104、17739、11984及14682条Unigenes成功注释。通过与KOG数据库进行比对,可分为25个不同的功能注释。与GO数据库进行比对,结果显示,共有33254条Unigene获得注释,这些功能注释分为三大类50个功能亚类。其中,生物过程最多。以KEGG数据库参考,共有11984条Unigenes参与133条代谢途径分支,以代谢相关的通路较为集中,找到了与花青素合成关键酶的Unigenes。本研究极大地丰富了野百合的基因资源,为进一步开展野百合功能基因及分子标记育种等方面的研究提供了一定理论支持与依据。 The transcr iptome of Lilium brownii was sequenced using Illumina Hiseq^(TM) 4000.After de novo assembly,47605 unigene were obtained with the total length of 33972306 bp,an average length of 713 bp and N50 of 1204 bp.All the Unigenes were searched against Nr,Swiss-Prot,KEGG and COG databases,and 28104,17739,11948 and 14682 Unigenes were respectively annotated in those databases.In KOG database,the annotated Unigenes were assigned to 25 KOG categories.33254 Unigenes were assigned with 50 GO categories under the three main categories The number of Unigene assigned to biological processes were most,a total of 11984 Unigenes were mapped to 133 KEGG pathways,and the largest pathway group was metabolism.In addition,the genes involved in anthocyanin biosynthesis pathway were discovered.The research enriched the genetic resources of Lilium brownii,and will be provided useful theory and basis for the future researches on functional gene and molecular breeding and molecular marker.
