摘要
目的:研究三黄凝胶对大鼠耳郭痤疮模型血清金属蛋白酶-13(Matrix Metalloproteinases-13,MMP-13)、γ-干扰素(Interferon-gamma,IFN-γ)含量的影响,讨论三黄凝胶治疗痤疮的作用机制。方法:把60只6~8周龄的Wistar大鼠采用随机数字表法分为6组,每组10只,任选其中一组为空白组,其余5组在大鼠左侧耳郭内导管开口处2.5 cm×2.5 cm涂抹100%油酸建模,持续抹药3周。肉眼和组织病理学观察造模成功后,除空白组外,其余5组随机分为模型组、阳性对照组、三黄凝胶高剂量组、三黄凝胶中剂量组、三黄凝胶低剂量组。实验第22天开始外用药物,空白组和模型组不给任何药,阳性对照组涂抹0.025%维A酸乳膏(剂量:5g/kg);三黄凝胶低剂量组、中剂量组、高剂量组:分别给予0.02 g、0.04 g、0.08 g生药/(kg•d),治疗28 d后采用ELISA法检测各组大鼠血清中MMP-13、IFN-γ的含量。结果:与空白组相比,模型组血清中MMP-13升高(P<0.01),IFN-γ降低(P<0.01),差异有统计学意义;各治疗组与模型组相比,血清中MMP-13水平下降(P<0.01),IFN-γ水平升高(P<0.01),差异有统计学意义;阳性对照组与三黄凝胶低剂量组、中剂量组相比,差异无统计学意义(P>0.05),三黄凝胶高剂量组与阳性对照组相比,血清中MMP-13水平降低(P<0.05),IFN-γ水平升高(P<0.05),差异有统计学意义。结论:三黄凝胶可通过减低血清中MMP-13的含量、升高IFN-γ的含量来达到治疗痤疮的目的。
Objective:To study the effect of Sanhuang gel(三黄凝胶)on serum metalloproteinase-13(MMP-13)and shapinterferon(IFN-γ)content in the rat model of auricle acne,and to discuss the mechanism of Sanhuang gel in the treatment of acne.Methods:Sixty Wistar rats aged from 6 to 8 weeks were randomly divided into the blank group,the model group,the positive control group,Sanhuang gel high-dose group,medium-dose group and low-dose group,with 10 rats in each group.100%oleic acid was applied to the opening of the catheter in the left auricle of rats for modeling(except the blank group).After successful modeling,all groups,except the blank group and model group,were given the drug for 28 days,and then the serum levels of MMP-13 and IFN-γin each group were detected by ELISA.Results:Compared with the model group,serum MMP-13 levels of the model group decreased(P<0.01),while IFN-γserum levels increased(P<0.01),showing statistically significant differences.Compared with the positive control group,the serum MMP-13 level of the low-dose group and the medium-dose group decreased(P<0.05),while the IFN-γlevel increased(P<0.05),showing statistically significant differences.Conclusion:Sanhuang gel can reduce the content of MMP-13 and increase the content of IFN-γto achieve the purpose of acne treatment.
