环鸟苷酸腺苷酸合成酶-干扰素基因刺激因子信号通路在肝脏疾病中作用研究进展 被引量:1

Research progress of cyclic GMP-AMP synthase-stimulator of interferon genes signaling pathway in liver diseases

摘要 环鸟苷酸腺苷酸合成酶-干扰素基因刺激因子(cGAS-STING)信号通路是内外源性异常核酸物质激活固有免疫系统的重要信号通路。cGAS-STING通路通过干扰素调节因子3(IRF3)或核因子-κB(NF-κB)活化促进Ⅰ型干扰素和炎性因子分泌介导免疫反应。研究结果表明cGAS-STING通路在炎症、感染及肿瘤发生发展中的作用越来越重要。适度激活cGAS-STING通路发挥防御作用,而过度激活cGAS-STING通路导致有害的炎性反应。同样,cGAS-STING通路在病毒性肝炎、肝癌、缺血再灌注损伤等肝脏常见疾病中起重要作用。cGAS-STING通路的发现为研究肝脏疾病的发生发展机制及探索更为有效的治疗方法提供了新的策略。本文结合现阶段cGAS-STING通路的研究进展,总结cGAS-STING通路在常见肝脏疾病领域的研究进展。 The cyclic GMP-AMP synthase-stimulator of interferon genes(cGAS-STING)signaling pathway is important for endogenous and external abnormal nucleic acid to activate the innate immune system.Via the activation of downstream signals such as interferon regulatory factor 3(IRF3)or nuclear factor-κB(NF-κB),the cGAS-STING pathway promotes the secretion ofⅠ-interferon and inflammatory factors to mediate immune responses.Recent studies have shown that cGAS-STING pathway plays an increasingly important role in inflammation,infection and carcinogenesis.Moderate activation of cGAS-STING pathway plays an immune defense role,but excessive activation of the cGAS-STING pathway leads to deteriorative inflammatory responses.Similarly,cGAS-STING pathway also plays an important role in liver diseases such as viral hepatitis,liver cancer and ischemia/reperfusion injury.The discovery of cGAS-STING pathway provides a new strategy for studying the pathogenesis of liver diseases and exploring more effective treatment methods.This paper is based on the current progress of cGAS-STING pathway to summarize the progress of cGAS-STING pathway in common liver diseases such as viral hepatitis,liver cancer and metabolic liver disease.
