真核起始因子4A及组蛋白乙酰转移酶表达水平与卵巢癌患者FIGO分期的相关性

Correlations Between the Expression Levels of Eukaryotic Initiation Factor 4A and Histone Acetyltransferase and FIGO Staging in Patients with Ovarian Cancer

摘要 目的分析真核起始因子4A(eIF4A)及组蛋白乙酰转移酶(HAT)表达水平与卵巢癌患者国际妇产科协会(FIGO)分期的相关性。方法选取2012年12月至2014年10月郑州人民医院收治的43例卵巢癌患者作为观察组,另选取同期40例良性卵巢疾病患者作为对照组。检测两组患者HAT、eIF4A表达情况,比较两组HAT及eIF4A阳性表达率、不同FIGO分期HAT及eIF4A阳性表达率以及HAT及eIF4A阳性表达率与FIGO分期的相关性,分析HAT、eIF4A表达水平与卵巢癌患者中位生存期的关系,采用多因素logistic回归分析影响卵巢癌患者预后的因素。结果观察组HAT阳性表达率低于对照组,eIF4A阳性表达率高于对照组(P<0.05)。FIGO分期Ⅰ~Ⅱ期HAT阳性表达率高于Ⅲ~Ⅳ期,eIF4A阳性表达率低于Ⅲ~Ⅳ期(P<0.05)。Spearman秩相关分析表明,HAT阳性表达率与FIGO分期呈负相关(r=-0.305,P<0.001),eIF4A阳性表达率与FIGO分期呈正相关(r=0.373,P<0.001)。HAT阴性表达患者中位生存期较阳性表达患者短,eIF4A阳性表达患者中位生存期较阴性表达患者短(P<0.05)。多因素logistic回归分析结果显示,HAT阴性、eIF4A阳性是卵巢癌患者中位生存期短的独立危险因素(P<0.05)。结论HAT及eIF4A在卵巢癌患者中存在异常表达,与卵巢癌发生、进展关系密切,HAT阴性、eIF4A阳性提示卵巢癌患者预后较差,检测HAT及eIF4A表达水平有助于卵巢癌早期诊断及预后分析,可为临床干预提供指导意见。 Objective To analyze the correlations between the expression levels of eukaryotic initiation factor 4A(eIF4A)and histone acetyltransferase(HAT)and Federation International of Gynecology and Obstetrics(FIGO)staging in patients with ovarian cancer.Methods A total of 43 patients with ovarian cancer in Zhengzhou People’s Hospital from December 2012 to October 2014 were selected as observation group,and 40 patients with benign ovarian disease during the same period were selected as control group.The expression levels of HAT and eIF4A in the two groups were detected.The positive expression rates of HAT and eIF4A in the two groups were compared.The positive expression rates of HAT and eIF4A in different FIGO stages and correlations between the positive expression rates of HAT and eIF4A and FIGO staging were compared between the two groups.The relationships between the expression levels of HAT and eIF4A and median survival time of patients with ovarian cancer were analyzed.Multivariate logistic regression analysis was used to analyze the prognostic factors of patients with ovarian cancer.Results The positive expression rate of HAT in observation group was lower than that in control group,and the positive expression rate of eIF4A in observation group was higher than that in control group(P<0.05).The positive expression rate of HAT in FIGO stageⅠtoⅡwas higher than that in stageⅢtoⅣ,and the positive expression rate of eIF4A in FIGO stageⅠtoⅡwas lower than that in stageⅢtoⅣ(P<0.05).Spearman rank correlation analysis showed that the positive expression rate of HAT was negatively correlated with FIGO staging(r=-0.305,P<0.001),and the positive expression rate of eIF4A was positively correlated with FIGO staging(r=0.373,P<0.001).The median survival time of negative expression of HAT was shorter than that of positive expression,and the median survival time of positive expression of eIF4A was shorter than that of negative expression(P<0.05).Multivariate logistic regression analysis showed that negative HAT and positive eIF4A were independent risk factors of short median survival in patients with ovarian cancer(P<0.05).Conclusion The abnormal expression of HAT and eIF4A is closely related to the occurrence and progression of ovarian cancer.The negative HAT and positive eIF4A indicate that patients with ovarian cancer have poor prognoses.The detection of the expression levels of HAT and eIF4A is helpful for early diagnosis and prognostic analysis of ovarian cancer,which can provide guidance for clinical intervention.
