摘要
目的:探讨胆碱激酶α(CHKA)抑制剂MN58b对U87胶质瘤干细胞(GSCs)增殖及凋亡的影响。方法:从U87细胞中分离肿瘤干细胞U87GSCs,并进行验证;检测U87MG、U87GSCS及小胶质细胞HMC3中CHKA表达。CHKA抑制剂MN58b作用于U87GSCs,检测CHKA生物活性减弱对U87GSCs增殖和凋亡的影响。结果:分离的U87GSCs可表达肿瘤干细胞和神经干细胞表面分子标志物CD133、Nestin;U87MG、U87GSCS中CHKA基因及蛋白表达明显高于小胶质细胞HMC3;CHKA抑制剂MN58b可有效抑制U87GSCs增殖,促进其凋亡。结论:CHKA可能是一种促癌基因,与GSCs相关,并影响胶质瘤发生发展。
Objective:To investigate effect of Choline Kinase α(CHKA)inhibitor MN58 b on proliferation and apoptosis of U87 glioma stem cells(GSCs).Methods:U87 GSCs tumor stem cells were isolated from U87 cells,and were verified. Expression of CHKA in U87 MG,U87 GSCS,and microglia HMC3 was detected. CHKA inhibitor MN58 b was applied to U87 GSCs to detect biological activity of CHKA weakening on proliferation and apoptosis of U87 GSCs.Results:Isolated U87 GSCs can express molecular markers CD133 and Nestin on surface of tumor stem cells and neural stem cells. Expressions of CHKA gene and protein in U87 MG and U87 GSCS were significantly higher than that of microglia HMC3. CHKA inhibitor MN58 b can effectively inhibit U87 GSCs proliferation ability and promote apoptosis ability.Conclusion:CHKA may be a cancer-promoting gene related to GSCs and affect occurrence and development of glioma.
