摘要
采用HPLC法结合BBD响应面法优化党参中党参炔苷的提取工艺并建立含量测定方法。以党参为原料,在单因素的基础上,采用Box-Behnken设计实验组别,选取超声时间、液料比和甲醇浓度作为影响因素,以党参中党参炔苷含量为指标,采用高效液相色谱法(HPLC)进行检测,确定党参中党参炔苷的最佳提取工艺。结果表明,党参中党参炔苷提取工艺的最佳条件为:超声提取时间为40 min,液料比为40,甲醇浓度为70%。在最佳条件下,党参炔苷的含量为0.152%。响应面分析法优化的含量测定提取工艺,参数准确可靠,提取效率高,方法可行。建立的含量测定方法稳定可靠,可为党参的质量控制体系建立提供理论基础和实用价值。
In this paper,HPLC and BBD response surface methodology were used to optimize the extraction process of lobetyolin from Codonopsis pilosula and a method for the determination of Lobetyolin was established.Taking Codonopsis pilosula as raw material,on the basis of single factor,the experimental group was designed by box-Behnken.The ultrasonic time,liquid-solid ratio and methanol concentration were selected as the influencing factors.The content of lobetyolin in Codonopsis pilosula was determined by high performance liquid chromatography(HPLC).The results showed that the optimum extraction conditions were as follows:ultrasonic extraction time was 40min,liquid-solid ratio was 40 and methanol concentration was 70%.Under the optimum conditions,the content of lobetyolin was 0.152%.The optimized extraction process of content determination by response surface analysis has the advantages of accurate and reliable parameters,high extraction efficiency and feasibility.The established method for determination of content is stable and reliable,which can provide theoretical basis and practical value for the establishment of quality control system of Codonopsis pilosula.
