摘要
目的探讨大鼠减体积自体肝移植模型的建立。方法20例SD大鼠在异氟烷吸入麻醉后,经门静脉肝脏原位冷灌注含有肝素钠(50 IU/ml)的乳酸钠林格液并切除肝中叶及左外叶,模拟临床自体肝移植手术过程,建立大鼠减体积自体肝移植模型。观察大鼠围手术期相关指标及存活情况。结果大鼠减体积自体肝移植模型20例,成功完成手术18例,1例术中死于分离肝上下腔静脉时损伤膈肌造成气胸,1例死于术后复苏体温过低而无法苏醒。大鼠平均麻醉诱导时间(1.9±0.3)min,手术时间(38.6±2.9)min,无肝期时间(16.0±0.9)min,切除肝重量(6.4±0.8)g,术后麻醉复苏时间(3.1±0.3)min。大鼠术后1周成活率94%(17/18)。结论大鼠减体积自体肝移植模型建立的操作相对简易,可重复性好,术后存活率高。该模型有效结合肝切除和原位肝移植技术,适合缺血-再灌注损伤和自体肝移植等机制研究。
Objective To investigate the establishment of rat models of reduced-size autologous liver transplantation.Methods After isoflurane inhalation anesthesia,20 SD rats were treated with cold perfusion in situ of sodium lactate Ringer's solution containing heparin sodium(50 IU/ml)via the portal vein,and then the middle and left lateral liver lobes were resected to simulate the clinical process of autologous liver transplantation.Relevant parameters and survival of rats during the perioperative period were observed.Results Among the 20 rats receiving reduced-size autologous liver transplantation,18 successfully survived.1 rat died of pneumothorax caused by diaphragm injury when separating the superior and inferior vena cava of the liver intraoperatively,and 1 rat died of hypothermia after postoperative resuscitation.The mean anesthesia induction time,operation time,anhepatic time,resected liver mass and postoperative recovery time from anesthesia were(1.9±0.3)min,(38.6±2.9)min,(16.0±0.9)min,(6.4±0.8)g and(3.1±0.3)min,respectively.The survival rate of rats at postoperative 1 week was 94%(17/18).Conclusions It is relatively convenient and reproducible to establish the rat models of reduced-size autologous liver transplantation,which yields high postoperative survival rate.This rat model effectively combines liver resection and orthotopic liver transplantation.
