SLC26A3在结直肠癌中的表达、功能及影响机制探讨

摘要 目的为分析SLC26A3在结肠癌中的表达、功能和PI3K/AKT信号通路对SLC26A3表达的影响。方法通过对数据库信息挖掘,探索SLC26A3在肿瘤疾病尤其是结直肠腺癌中的表达情况,分析其在结直肠癌中可能发挥的功能。利用细胞学实验,观察分析PI3K/AKT信号通路对SLC26A3表达的影响。结果SLC26A3在肿瘤组织中的表达相较正常组织呈现低水平表达。虽未发现SLC26A3对于结直肠腺癌的肿瘤分期和预后有显著影响,但高表达SLC26A3的病人长期生存的发生率较高。在Caco-2细胞中,激活PI3K/AKT信号通路低激活状态对SLC26A3的表达起促进作用,但持续过度激活对SLC26A3的表达起抑制作用(P<0.05)。结论SLC26A3在结直肠癌中低表达,SLC26A3高表达有利于患者的长期生存,PI3K/AKT信号通路对SLC26A3的表达可能发挥着双向调节的作用。 Objective We aimed to explore the expression and function of SLC26A3 and the influence of PI3K/AKT pathway on its expression in colorectal cancer(CRC).Methods By mining the public database,the expression of SLC26A3 in cancers,especially colorectal adenocarcinoma(CRA)was explored and its potential functions in CRC were predicted.What’s more,based on cytology experiments,the effects of PI3K/AKT signaling pathway on the expression of SLC26A3 were analyzed.Results The expression of SLC26A3 in cancerous tissues was lower than that in normal tissues.Meanwhile,the expression of SLC26A3 has not been found to be significantly associated with tumor staging and prognosis of CRA patients.In Caco-2 cells,activation of AKT signaling pathway promotes the expression of SLC26A3,but its continuous activation inhibits the expression of SLC26A3(P<0.05).Conclusion SLC26A3 is low expressed in CRC and its high expression is beneficial to long-term survival.PI3K/AKT signaling pathway might bidirectionally regulate the expression of SLC26A3,which might further impact its functions.
