摘要
铁死亡是最近被定义的一种铁依赖性的,由脂质过氧化物累积所触发的,不同于坏死、凋亡、自噬的细胞调节性死亡(regulated cell death,RCD)方式。铁死亡与肿瘤密切相关,有望成为治疗肿瘤的一个新的突破点。同时,肿瘤细胞可通过抑制铁死亡,增强活性氧(reactive oxygen species,ROS)失衡介导的氧化应激防御能力,诱导肿瘤细胞耐药。本研究对铁死亡的发生机制以及铁死亡在肿瘤耐药中发挥的作用进行综述,为克服肿瘤耐药提供新的思路。
Ferroptosis is a recently defined iron-dependent,triggered by the accumulation of lipid peroxides,which is different from the regulated cell death(RCD)mode of necrosis,apoptosis,and autophagy.Ferroptosis is closely related to tumors and is expected to become a new breakthrough point in the treatment of tumors.At the same time,tumor cells can inhibit iron death,enhance the oxidative stress defense ability mediated by reactive oxygen species(ROS)imbalance,and induce tumor cell resistance.This study reviewed the mechanism of ferroptosis and the role of ferroptosis in tumor resistance,and provided new ideas for overcoming tumor resistance.
