摘要
目的建立HPLC法测定金荞麦药材中表儿茶素、金丝桃苷、槲皮苷的分析方法。方法采用Waters C18色谱柱(4.6mm×150mm,5μm),表儿茶素测定的流动相为乙腈-0.004%磷酸水溶液(10:90,v/v),检测波长为280nm;金丝桃苷测定的流动相为甲醇-0.004%磷酸水溶液(50:50,v/v),检测波长为345nm;槲皮苷测定的流动相为甲醇-0.004%磷酸水溶液(45:55,v/v),检测波长为345nm。流速均为1.OmL/min;柱温均为35℃;进样量均为10μL。结果表儿茶素、金丝桃苷、槲皮苷在0.40~2.00μg范围内与峰面积积分值呈良好的线性关系,平均加样回收率分别为99.5% (RSD=2.73%)、99.14%(RSD=0.12%)、98.7%(RSD=1.98%)。结论该方法操作简便,准确可靠,重复性好,专属性强,并且分析时间短,可作为金荞麦的质量控制的有效手段。
Objective An HPLC method for the determination of epicatechin,hyperoside and quercitrin from Fagopyri Dibotryis Rhizomam was established in this paper.Methods the Waters C18 column(4.6 mmx150 mm,5.0μm) was adopted to separate the samples.acetonitrile-0.004% phosphoric acid solution,methanol-0.004% phosphoric acid solution was used as a mobile phase at a flow of 1.0 mL/min.the column temperature was 35℃,and the detection wavelength was 280,345,345 nm in the order,the sample volume was 10 μL.Results The three active components showed good linear relationship respectively over the concentration range of 0.40~2.00μg,the average recovery was 99.5%(RSD=2.73%)、99.14%(RSD=0.12%)、98.7%(RSD=1.98%).Conclusion The establishment of the determination of indicators is superior to the existing pharmacopoeia determination method,can be used as Fagopyri Dibotryis Rhizomam content determination method.
