丹参饮预处理对大鼠心肌缺血再灌注损伤的保护作用研究 被引量:4

摘要 目的从炎症角度研究丹参饮预处理对大鼠心肌缺血再灌注损伤的保护作用。方法将50只SD大鼠随机分为假手术组、模型组、丹参饮低剂量组、丹参饮中剂量组、丹参饮高剂量组,每组10只。假手术组和模型组给予生理盐水灌胃,丹参饮低、中、高剂量组分别给予丹参饮5 g(生药量)/kg、10 g(生药量)/kg、15 g(生药量)/kg灌胃,均连续14 d。第15天,除假手术组外,其余组均采用前降支结扎法制备大鼠心肌缺血再灌注损伤模型。记录各造模大鼠缺血再灌注过程中心电图;再灌注3 h后腹主动脉取血,检测各组大鼠血清肌酸激酶同工酶(CK-MB)、乳酸脱氢酶(LDH)、高敏C反应蛋白(hs-CRP)水平;取缺血心肌组织,HE染色进行心肌病理学观察。结果前降支结扎即刻心电图表现为ST段呈弓背向上抬高;前降支再灌注后出现室性心律失常;再灌注180 min后ST段回落,病理性Q波形成。模型组大鼠血清CK-MB、LDH、hs-CRP水平均明显高于假手术组(P均<0.05);丹参饮各组大鼠血清CK-MB、LDH、hs-CRP水平均明显低于模型组(P均<0.05),且丹参饮中、高剂量组均明显低于丹参饮低剂量组(P均<0.05),丹参饮高剂量组均明显低于丹参饮中剂量组(P均<0.05)。HE染色见模型组大鼠心肌纹理紊乱,心肌细胞结构破坏;丹参饮各组心肌损伤较模型组轻,丹参饮高剂量组最轻。结论丹参饮预处理可能通过抑制炎症反应,减轻心肌细胞缺血再灌注损伤,保护缺血心肌,且存在剂量依赖关系。 Objective It is to study the protective effect of pretreatment with Danshen Decoction on myocardial ischemia/reperfusion injury in rats from the perspective of inflammation.Methods Fifty SD rats were randomly divided into 5 groups:sham operation group,model group,low,medium and high dose Danshen Decoction groups,10 rats in each group.The sham operation group and the model group were given normal saline by gavage,and the low,medium and high-dose Danshen Decoction groups were respectively given 5 g(crude drug)/kg,10 g(crude drug)/kg,15 g(crude drug)/kg Danshen Decoction.All the groups were continuously treated for 14 days.On the 15th day,except for the sham operation group,the rest groups were all used to prepare rat myocardial ischemia-reperfusion injury models by ligation of the anterior descending branch.The electrocardiogram of the ischemia-reperfusion process of the rats in each group was recorded;the blood was taken from the abdominal aorta after 3 hours of reperfusion,and the levels of serum creatine kinase isoenzyme(CK-MB),lactate dehydrogenase(LDH),high-sensitivity C-reactive protein(hs-CRP)were detected;the ischemic myocardial tissue was taken and stained with HE for myocardial pathology observation.Results Immediately after ligation of the anterior descending branch,the ECG showed that the ST segment was arched upward;the ventricular arrhythmia occurred after reperfusion of the anterior descending branch;the ST segment fell after 180 minutes of reperfusion,and pathological Q waves formed.The levels of serum CK-MB,LDH,and hs-CRP in the model group were significantly higher than those in the sham operation group(all P<0.05);the levels of serum CK-MB,LDH,and hs-CRP in the Danshen Decoction groups were significantly lower than those in the model group(all P<0.05),and the medium and high dose of Danshen Decoction groups were significantly lower than the low dose of Danshen Decoction group(P<0.05),and the high dose group was significantly lower than the medium dose group(all P<0.05).HE staining showed that the myocardial texture of rats in the model group was disordered,and the structure of myocardial cells was damaged;the myocardial damage in each group of Danshen Decoction was lighter than that of the model group,and the high-dose Danshen Decoction group was the lightest.Conclusion Pretreatment with Danshen Decoction can relieve myocardial cell ischemia/reperfusion injury and protect the ischemic myocardium by inhibiting inflammatory response,its effect is dose dependent.
