摘要
目的利用micro-CT及双能X线吸收测量仪(dual energy X-ray absorptiometry,DXA)观察Piezo1蛋白对去卵巢小鼠骨量及骨微结构的影响。方法选取5只雌性Piezo1ΔEC小鼠为实验组,模型组和对照组各选取5只雌性Piezo1fl/fl小鼠,实验组与模型组均行去卵巢手术后于14周龄时使用micro-CT和DXA检测和分析各组小鼠胫骨的骨密度(bone mineral density,BMD)、骨矿物质含量(bone mineral content,BMC)、骨小梁相对体积(bone volume/tissue volume,BV/TV)、骨小梁厚度(trabecular bone thickness,Tb.Th)、骨小梁数量(trabecular bone number,Tb.N)、骨小梁分离度(trabecular bone separation,Tb.Sp)、骨结构模型指数(structure model index,SMI)及骨皮质厚度(cortical bone thickness,Ct.Th),然后使用SPSS软件进行统计学分析。结果DXA结果显示实验组BMC值明显低于模型组,实验组:(0.003±0.000)g;模型组:(0.005±0.001)g(P<0.05)。Micro-CT结果显示实验组骨质疏松主要体现在骨小梁数量减少和骨小梁分离度增加,其中实验组和模型组骨小梁数量分别为0.549±0.013和0.631±0.015;骨小梁分离度分别为0.453±0.018和0.362±0.046(P<0.05)。结论内皮细胞Piezo1缺失能增加去势小鼠的骨质疏松严重程度。
Objective To observe the effects of Piezo1 on bone mass and bone microstructure in ovariectomized mice by micro-CT and dual energy X-ray absorptiometry(DXA).Methods Five female Piezo1ΔECmice were selected as the experimental group.Model group and control group contained 5 female Piezo1fl/fl mice,separately.Ovariectomized mouse were performed in the experimental group and the model group.When those mice were 14 weeks old,we used micro-CT and DXA to test and analyze of the type of tibia bone mineral density,bone mineral content(BMC),trabecular bone volume,the bone trabecular thickness,number of trabecular bone(Tb.N),bone trabecular separation degree(Tb.Sp),structure model index and cortical bone thickness of tibia.ResultsThe outcomes of DXA showed that BMC value in the experimental group was significantly lower than that in the model group.Micro-CT results showed that the number of bone trabeculae decreased and the separation of bone trabeculae increased in the experimental group.Tb.N in the experimental group and the model group were 0.549±0.013 and 0.631±0.015,respectively.Tb.Sp in the experimental group and the model group were 0.453±0.018 and 0.362±0.046(P<0.05).Conclusion The Piezo1 deficit in endothelial cells increased the severity of osteoporosis in ovariectomized mice.
