miRNA-2861、miRNA-483-3p与多囊卵巢综合征患者内分泌代谢及卵母细胞质量的相关性 被引量:5

Correlation between miRNA-2861,miRNA-483-3p and endocrine metabolism and oocyte quality in patients with polycystic ovary syndrome

摘要 目的:探讨miRNA-2861、miRNA-483-3p在多囊卵巢综合征(PCOS)患者血清和颗粒细胞中的差异表达,及其对体外受精(IVF)过程中卵母细胞质量及胚胎发育潜能的影响。方法:收集2016年4月至2018年1月于山东中医药大学附属医院生殖中心行显微受精-胚胎移植(ICSI)的PCOS患者58例(PCOS组)和单纯因男方因素不孕的患者23例(对照组)。RT-PCR法检测PCOS组与对照组血清及颗粒细胞中miR-2861、miR-483-3p的表达水平。结果:miR-2861、miR-483-3p在血清和颗粒细胞中表达水平具有一致性,PCOS组均低于对照组,差异有统计学意义(P<0.05)。PCOS组血清中miR-2861表达水平与空腹胰岛素、总胆固醇呈负相关,PCOS组血清中miR-483-3p表达水平与睾酮、空腹胰岛素、低密度脂蛋白呈负相关(P<0.05)。PCOS组颗粒细胞中miR-2861表达水平与获卵率、MII卵率、2PN卵裂率、优质胚胎率呈正相关(P<0.05)。PCOS组颗粒细胞中miR-483-3p表达水平与获卵率、MII卵率、正常受精率、2PN卵裂率、优质胚胎率呈正相关(P<0.05)。结论:miR-2861和miR-483-3p在PCOS患者血清、颗粒细胞中异常低表达,异常表达的miR-2861、miR-483-3p可能使卵母细胞及胚胎质量下降,间接影响IVF成功率。 Objective:To investigate the differential expression of miRNA-2861 and miRNA-483-3p in serum and granulosa cells in PCOS patients,and its effect on oocyte quality and embryo development potential during in vitro fertilization(IVF).Methods:58 PCOS patients underwent ICSI from April 2016 to January 2018 were enrolled as the study group(PCOS group),while 23 infertile patients with male factors as a control group.The expression levels of miR-2861 and miR-483-3p in serum and granulosa cells of all groups were detected by RT-PCR.Results:The expression levels of miR-2861,miR-483-3p in serum and granulosa cells were lower in the PCOS group than those in the control group.The difference was significant(P<0.05).The expression level of miR-2861 in serum of PCOS group was correlated with fasting insulin and total cholesterol negatively.The expression level of miR-483-3p in serum of PCOS group was correlated with testosterone,fasting insulin and low density lipoprotein negatively(P<0.05).The level of miR-2861 in granulosa cells of PCOS group was positively correlated with egg capture rate,MII egg rate,2PN cleavage rate and high quality embryo rate(P<0.05).The level of miR-483-3p in granulosa cells of PCOS group was positively correlated with egg capture rate,MII egg rate,normal fertilization rate,2PN cleavage rate and high quality embryo rate(P<0.05).Conclusion:The expressions of miR-2861 and miR-483-3p are significant lower in serum and granulosa cells of PCOS patients than those of control patients,and they may decrease the quality of oocytes and embryo,indirectly affecting IVF success rate of PCOS patients.
