分析超速离心技术在蛋白质研究中的应用

摘要 分析超速离心技术(analytical ultracentrifugation,AUC)是检测生物大分子溶液中生物物理性质的重要技术,利用沉降速率(sedimentation velocity,SV)和沉降平衡(sedimentation equilibrium,SE)方法,通过紫外/可见光、干涉光以及荧光检测器来记录样品池中生物大分子径向浓度分布,以获得其水力学、生物物理学特性。该文介绍了分析超速离心技术的产生与发展、测试原理与数据分析方法,列举了分析超速离心技术在蛋白质性质分析、蛋白质与生物分子间相互作用测定、蛋白质结构解析以及蛋白类生物类似药分析中的应用实例,并展望了其在蛋白质研究中的应用与发展前景。 Analytical ultracentrifugation(AUC)is an important technique for the detection of biophysical properties in biomacromolecule solutions.the radial concentration distribution of biomacromolecules in the sample pool is recorded by ultraviolet/visible light,interference light and fluorescence detector to obtain its hydraulic and biophysical characteristics by using sedimentation velocity(SV)and sedimentation equilibrium(SE).This paper introduces the origin and development of analytical ultracentrifugation technology,test principle and data analysis method,lists the application examples of analytical ultracentrifugation technology in protein property analysis,determination of protein biomolecular interaction,protein structure analysis and analysis of protein biological analogues,and looks forward to its application and development prospect in protein research.
