摘要 目的采用高效液相色谱测定五维赖氨酸颗粒中维生素B6、维生素B2、维生素B1、烟酰胺、泛酸钙的含量。方法色谱柱为DIONEX C18柱(5μm,4.6 mm×250 mm);色谱柱温度为30℃;流动相为三乙胺-三氟乙酸-甲醇-0.02 mmol/L庚烷磺酸钠溶液(2∶3∶220∶775),检测器为二极管阵列检测器(PDA);检测波长为270 nm(维生素B6、维生素B2、维生素B1、烟酰胺)和220 nm(泛酸钙)。采用配置PDA检测器的高效液相色谱仪测定五维赖氨酸颗粒中5种成分的含量。结果供试品溶液色谱图中在与对照品混合溶液色谱图相应的时间位置有相同的色谱峰,阴性样品溶液无干扰。5种化合物在30 min内实现分离,分离度均在1.5以上,并且在一定浓度范围内呈现良好的线性关系,维生素B6、维生素B2、维生素B1、烟酰胺、泛酸钙在各自的线性范围内的相关系数(r)为0.991~0.999,检出限为1.25~18.0μg/mL。结论高效液相色谱法具有精密度好、简捷、高效的特点,可以作为五维赖氨酸颗粒中几种成分的含量测定方法。 Objective To determine the contents of vitamin B6,vitamin B2,vitamin B1,nicotinamide and calcium pantothenate in five vitamins and lysine granules by high performance liquid chromatography.Methods The chromatographic column was DIONEX C18 column(5μm,4.6 mm×250 mm);the column temperature was 30℃;the mobile phase was triethylamine-trifluoroacetic acid-methanol-0.02 mmol/L sodium heptanesulfonate solution(2∶3∶220∶775),and the detector was photodiode array(PDA);the detection wave lengths were 270 nm(vitamin B6,vitamin B2,vitamin B1,nicotinamide)and 220 nm(calcium pantothenate).The contents of five components in five vitamins and lysine granules were determined by high performance liquid chromatography equipped with PDA detector.Results The chromatogram of the test solution has the same chromatographic peak at the time position corresponding to the chromatogram of the mixed solution of the control,while the negative sample solution has no interference.The five compounds separated in 30 minutes,the resolution was above 1.5,and showed a good linear relationship in a certain concentration range.The correlation coefficient(r)of vitamin B6,vitamin B2,vitamin B1,nicotinamide and calcium pantothenate in their respective linear range was 0.991-0.999,and the detection limit was 1.25-18.0μg/mL.Conclusion High performance liquid chromatography has the characteristics of good precision,simplicity and high efficiency.It can be used for the determination of contents of several components in five vitamins and lysine granules.
