乌梅丸及其拆方对TNBS致溃疡性结肠炎大鼠的作用研究 被引量:8

摘要 目的:基于2,4,6-三硝基苯磺酸(2,4,6-trinitrobenzenesulfonic acid,TNBS)诱导的大鼠溃疡性结肠炎(ulcerative colitis,UC)模型探索乌梅丸中酸、苦、甘、辛4类药物对UC大鼠的治疗作用及相互关系。方法:将乌梅丸按药味分成酸、苦、甘和辛4个单因素,使用L 8(27)正交表设计实验分组,采用TNBS法制备大鼠UC模型,观察给药后大鼠的结肠质量长度比、结肠组织中白介素(interleukin,IL)-1β和环氧化酶(cyclooxygenase,COX)-2的含量。结果:与正常组比较,模型组大鼠结肠质量长度比显著增大,IL-1β和COX-2含量均显著升高(P<0.05),差异有统计学意义。酸、辛和甘味药物对大鼠结肠质量长度比具有显著影响(酸>辛>甘),全方组、酸+甘组和甘+辛组可显著降低UC大鼠结肠质量长度比(P<0.05),差异均有统计学意义;苦味药物对IL-1β含量具有显著负影响(P<0.05),差异有统计学意义。酸和甘味药物对COX-2含量具有显著性影响(酸>甘),酸+甘药物组合对UC大鼠结肠组织的COX-2含量有显著改善作用(P<0.01),差异均有统计学意义。结论:乌梅丸及其各拆方组对TNBS诱导的大鼠UC模型具有显著调节作用,在乌梅丸对于UC的应用治疗中,酸、甘和辛味药物配伍起主导作用,苦味药物也具有重要配伍作用。 Objective:Based on the 2,4,6-trinitrobenzenesulfonic acid(TNBS)-induced ulcerative colitis(UC)model in rats,to explore the therapeutic effects and mutual relationship of sour,bitter,sweet and spicy medicines in Wumei pills on UC rats.Methods:According to the medicinal taste,Wumei Pill was divided into sour,bitter,sweet and spicy medicine.L 8(27)orthogonal table was used to design experiment.The rat model of UC was prepared by TNBS.Then the mass length ratio of colon and the contents of cytokines interleukin(IL)-1βand cyclooxygenase(COX)-2 in colon tissue were observed after administration.Results:Compared with the normal group,the colon mass length ratio,the contents of IL-1βand COX-2 in the model group increased significantly(P<0.05).The difference was statistically significant.Sour,spicy and sweet medicines had significant effects on the colonic mass-length ratio(sour>spicy>sweet).The whole prescription group,sour+sweet group and sweet+spicy group could significantly reduce the ratio of colon mass length in UC rats(P<0.05).The difference was statistically significant.Bitter medicine had a significant negative effect on IL-1β(P<0.05).The difference was statistically significant.Sour and sweet medicines had significant effect on COX-2(sour>sweet).Sour and sweet group had a significant regulatory effect on COX-2 in the colon tissue of UC rats(P<0.05).The difference was statistically significant.From the above results,sour+sweet+spicy group was the best combination of medicines for Wumei Pill to treat UC.Conclusion:Wumei Pill and its decomposed prescription groups can significantly regulate the UC model induced by TNBS in rats.In the application and treatment of Wumei Pill for UC,the compatibility of acid,sweet and spicy medicines played a leading role,and bitter medicine also played an important role in compatibility.
