双花保健口服液的制备及对应激性肝损伤的保护作用

Preparation of double flower health oral liquid and its protective effect on liver injury induced by stress

目的探索双花保健口服液的最佳制备工艺并对双花保健口服液对应激性肝损伤的保护作用进行药效学评价。方法得到金银花、菊花乙醇提取物,设计正交试验,探索最佳的双花保健口服液制备条件,并通过《中国药典》检测口服液是否符合质量标准。进一步利用拘束负荷的方法建立应激性肝损伤小鼠模型,依次对小鼠血清中谷丙转氨酶(ALT)、小鼠肝组织超氧化物歧化酶(SOD)的活力以及丙二醛(MDA)的含量进行测定,观察双花口服液对小鼠肝脏应激性肝损伤的影响,以评价双花保健口服液对应激性肝损伤的保护作用。结果正交试验得出双花口服液最佳的制备工艺为:8%双花浸膏混合物(金银花:菊花=1:1),4%蔗糖,0.10%柠檬酸。该工艺制得的口服液符合药典所规定的质量标准。小鼠拘束负荷应激实验结果表明,双花保健口服液对应激性肝损伤有一定的保护作用,能降低ALT水平,升高SOD活力,降低MDA的含量。结论正交试验优化出的双花保健口服液的最佳处方条件符合《中国药典》的质量标准,双花保健口服液对小鼠的应激性肝损伤有一定的保护作用。

Objective:To explore the best preparation process of the double flower health oral liquid and to evaluate the protective effect of the double flower health oral liquid on stress-induced liver injury.Methods:Important active components were extracted from ethanol extract of honeysuckle and chrysanthemum,the best conditions for the preparation of oral liquid oral liquidand suitable accessories were explored by designing the orthogonal experiment,and whether the oral liquid met the quality standards was detected.The effect of honeysuckle and chrysanthemum double flower oral liquid on liver stress induced liver injury in mice was further explored to evaluate the protective effect of the double flower health oral liquid on the induced liver injury.Establishment of stress liver injury mouse model by restraint load method,the activities of ALT in serum and the content of SOD and MDA in liver were determined to evaluate the hepatoprotective effects.Results:Orthogonal test showed that the best preparation technology of the double flower oral liquid was:8%the mixture of flower extract(honeysuckle'chrysanthemum=1*1),4%sucrose and 0.10%citric acid.The oral liquid prepared by this process accords with the quality standard stipulated in the Pharmacopoeia.The experimental results showed that the double flower health oral liquid had certain protective effects on induced liver injury,reduced the level of ALT,increased the level of SOD and reduced the content of MDA.Conclusion:Through orthogonal test,the best prescription condition of the double flower health oral liquid is optimized,which accords with the quality standard of Chinese Pharmacopoeia.At the same time,the double flower health oral liquid can protect and improve stress liver injury in mice.
