基于APN/AMPK信号通路探讨黄芪黄酮与葛根黄酮配伍对糖尿病大鼠骨骼肌糖脂代谢的调控作用 被引量:4

摘要 目的观察黄芪黄酮、葛根黄酮对糖尿病大鼠骨骼肌糖脂代谢的影响。方法 SPF级SD雄性大鼠86只,随机分为6组,除正常组外,其余各组均采用链脲佐菌素(STZ)诱导糖尿病模型。造模同时,黄芪黄酮组、葛根黄酮组、黄芪黄酮+葛根黄酮组分别按0.039 g/kg、0.036 g/kg、0.075 g/kg剂量灌胃给药;中药对照组用金芪降糖片混悬液1.47 g/kg;正常组、模型组给予等体积蒸馏水10 mL/kg,各组连续给药30 d。酶联免疫吸附试验法(ELISA)检测:脂联素(APN)、葡萄糖转运蛋白4(GLUT4);实时荧光定量聚合酶链式反应法(Real-time PCR)检测:脂联素受体1(AdipoR1)、腺苷酸活化蛋白激酶(AMPK)、胰岛素受体底物2(IRS2)和磷脂酰肌醇3激酶(PI3K)表达。结果与正常组比较,模型组大鼠骨骼肌APN、GLUT4水平及AdipoR1、AMPK、IRS2、PI3K表达降低(P<0.05)。与模型组比较,3个实验组均有明显的改善,且黄芪黄酮和葛根黄酮的配伍对于胆固醇(CHO)和甘油三酯(TG)调节存在交互作用。结论推测黄芪黄酮与葛根黄酮降低血糖、血脂作用,与其参与骨骼肌APN/AMPK的相关信号通路有关。 Objective To observe the effects of astragalus flavone and pueraria flavone on glucose and lipid metabolism in skeletal muscle of diabetic rats.Methods 86 male SD rats of SPF grade were randomly divided into 6 groups.Except the normal group, the other groups were induced by streptozotocin(STZ). At the same time, Astragalus flavone group, Pueraria flavone group, astragalus flavone and pueraria flavone group were given 0.039 g/kg, 0.036 g/kg and 0.075 g/kg respectively by gavage;the traditional Chinese medicine control group was given Jinqi Jiangtang Tablet suspension of 1.47 g/kg;the normal group and model group were given equal volume of distilled water 10 mL/kg, and each group was given medicine for 30 days. APN and GLUT-4 were detected by enzyme linked immunosorbent assay(ELISA), and AdipoR1, AMPK, IRS2 and PI3 K were detected by real-time PCR.Results Compared with the normal group, the levels of APN, GLUT4 and theexpression of AdipoR1, AMPK, IRS2 and PI3 K in the skeletal muscle of the model group were decreased(P<0.05).Compared with the model group, the three experimental groups were significantly improved, and the compatibility of astragalus flavone and pueraria flavone had interaction effect on the regulation of CHO and TG.Conclusion It is speculated that astragalus flavone and pueraria flavone can reduce blood glucose and blood lipid, which is related to their involvement in APN/AMPK signaling pathway in skeletal muscle.
