摘要 目的基于网络药理学探讨黄芪治疗糖尿病肾病(diabetic kidney disease, DKD)的作用机制。方法应用中药系统药理学数据库及分析平台(TCMSP)筛选出黄芪的活性成分及其作用靶点,采用OMIM数据库,Drugbank数据库,TTD数据库,GeneCards数据,检索与DKD相关的靶点基因。利用Venny在线软件筛选黄芪与DKD的相交基因集,筛选关键靶点构建"黄芪-活性成分-靶点-DKD",基于DAVID数据库对关键靶点进行GO功能富集和KEGG通路富集分析。结果黄芪有效活性成分16个,治疗DKD的关键活性成分4个,关键作用靶点37个;GO功能富集分析得到326个条目,生物过程262个条目,细胞组分19个条目、分子功能45个条目;KEGG通路富集分析得出96个条目,主要包括TNF信号通路、Toll样受体信号通路、NOD样受体信号通路等。结论研究结果体现了黄芪多成分、多靶点、多通路作用治疗DKD,为黄芪治疗DKD的药理作用研究和临床应用提供新的理论依据。 Objective To explore the mechanism of Astragalus membranaceus in treating diabetic kidney disease based on network pharmacology.Methods The active components of Astragalus membranaceus and their target sites were selected by using TCMSP, and the target genes related to DKD were searched by OMIM database, Druggbank database, TTD database and GeneCards database. The crossing gene sets of Astragalus membranaceus and DKD were screened by using the on-line Software Venny, and the "Astragalus membranaceus-active components-target-DKD" was constructed by Cytoscape 3.6.1 software. The key targets were enriched by GO function and KEGG pathway based on DAVID Database.Results There were 16 active components of Astragalus Membranaceus, corresponding to 199 target sites, 4 key active components of DKD and 37 key target sites, 326 items and 262 items of biological process were obtained by GO enrichment analysis, there were 19 items of cell component and 45 items of molecular function, and 96 items were obtained by KEGG pathway enrichment analysis, it mainly includes TNF signaling pathway, Toll-like receptor signaling pathway, NOD-like receptor signaling pathway and so on.Conclusion The mechanism of action of Astragalus in the treatment of diabetic kidney disease is discussed by network pharmacology analysis.The results show that the Astragalus membranaceus has many components, many targets and many pathways, which can interact to form a complex biological network system, this study provides a new theoretical basis for the pharmacological action and clinical application of Astragalus membranaceus in the treatment of DKD.
