摘要 目的探索通过高效空气过滤器(HEPA)作为室内空气采样器,分析大气污染期间室内空气微生物群落结构组成。方法选取6户使用同一品牌HEPA过滤器的家庭,采用Illumina Miseq平台16SrRNA测序技术对HEPA滤膜上空气微生物进行测序。结果通过Illumina Miseq测序,6个HEPA滤膜空气样品共获得451878个有效序列,主要微生物群落为厚壁菌门(Firmicutes)、放线菌门(Actinobacteria)、变形菌门(Proteobacteria)、拟杆菌门(Bacteroidetes)和蓝藻细菌(Cyanobacteria)。主坐标分析发现不同室内微生物群落结构差异较大。对16SrRNA测序结果分析发现大部分为非致病微生物,少数为条件致病菌。结论HEPA过滤器可作为大气污染期间室内空气长期采样器,反映室内空气微生物群落结构。 Objective To analyze the bacterial communities of indoor air via the membrane of HEPA filters during the haze days in Beijing.Methods Six families using the same brand of HEPA filters were selected and the membrane of HEPA filters used for one year were collected,microorganism of in the membrane of HEPA filters were sequenced by high-throughput sequencing of 16 SrRNA on the platform of Illumina Miseq.Results A total of 451878 genes sequences in six membranes of HEPA filters for purifying indoor air were obtained by Illumina Miseq sequencing,the most abundant phyla of which were Firmicutes,Actinobacteria,Proteobacteria,Bacteroidetes and Cyanobacteria respectively.The PCoA analyses showed that the microbiome diversity of indoor air had difference in the microbial flora structure,and it was also found that most microbial flora were nonpathogenic with a small portion of opportunistic pathogen.Conclusion HEPA filters may be used as an alternative air sampler during the haze days for a long period which can reflect the structure of bacterial community.
