丹参酮ⅡA磺酸钠对饮水砷暴露诱导小鼠肝脏毒性的拮抗作用 被引量:1

摘要 目的探究丹参酮ⅡA磺酸钠对亚砷酸钠诱导的小鼠肝脏毒性的拮抗作用,为地方性砷中毒的防治提供依据。方法将小鼠随机分为5个组,分别为对照组、丹参酮ⅡA磺酸钠(8 mg/kg)组、亚砷酸钠组、低剂量干预组(亚砷酸钠+丹参酮ⅡA磺酸钠8 mg/kg)、高剂量干预组(亚砷酸钠+丹参酮ⅡA磺酸钠20 mg/kg)。亚砷酸钠(50 mg/L)采用自由饮水的方式给予,丹参酮ⅡA磺酸钠腹腔注射给药,对照组腹腔注射葡萄糖溶剂及自由饮用去离子水;亚砷酸钠比丹参酮ⅡA磺酸钠提前预给药3天。测定小鼠血清中丙二醛(MDA)含量和总超氧化物歧化酶(T-SOD)的活力以及肝脏组织中酸性磷酸酶(ACP)、谷草转氨酶(AST)和乳酸脱氢酶(LDH)的活力。结果与亚砷酸钠染毒组相比,高剂量干预组血清中的MDA含量显著降低、T-SOD活力显著升高,差异均有统计学意义(P<0.05);高剂量干预组小鼠肝脏组织中LDH、AST活力与亚砷酸钠组相比都有所升高,其中LDH的升高具有统计学差异(P<0.05);而高剂量干预组的ACP活力与亚砷酸钠组相比有下降趋势,但无统计学意义(P>0.05)。结论丹参酮ⅡA磺酸钠可以增强小鼠肝脏的抗氧化能力,从而拮抗亚砷酸钠诱导的肝脏功能损害。 Objective To understand the antagonistic effects of tanshinone IIA sodium sulfonate on sodium arsenite-induced liver toxicity in mice,and to provide a basis for the prevention and treatment of endemic arsenism.Methods C57/BL-6 male mice were randomly divided into five groups,namely control group,sodium tanshinone IIA sulfonate(8 mg/kg)group,sodium arsenite group,low-dose intervention group(sodium arsenite+8 mg/kg tanshinone IIA sodium sulfonate)and high-dose intervention group(sodium arsenite+20 mg/kg tanshinone IIA sodium sulfonate).In the exposure groups,sodium arsenite(50 mg/L)was given through free drinking water three days before tanshinone IIA sulfonate was intraperitoneally injected;The control group was given glucose solvent through intraperitoneal injection and deionized water as the drinking water.The content of malondialdehyde(MDA)and the activity of total superoxide dismutase(T-SOD)in serum,the activities of acid phosphatase(ACP),aspartate aminotransferase(AST)and lactate dehydrogenase(LDH)in liver tissues of mice were determined.Results Compared with the sodium arsenite exposure group,significantly lower MDA content and significantly higher T-SOD activity in serum were observed in the high-dose intervention group(P<0.05);Higher activities of LDH and AST,lower activity of ACP in liver tissue were observed in high-dose intervention group,only LDH presented statistically significant difference(P<0.05).Conclusion Sodium tanshinone IIA sulfonate can enhance the antioxidant capacity in liver of mice,thereby antagonizing the liver injury induced by sodium arsenite.
