分离自红棕象甲虫尸上的绿僵菌菌株鉴定及其对红脉穗螟的致病力 被引量:1

摘要 红脉穗螟是海南省椰子、槟榔等作物的重要害虫,已引起了严重为害,造成了一定的经济损失。自死亡的红棕象甲虫分离得到一株绿僵菌菌株Red-3,通过形态特征和分子生物学方法对其进行了鉴定,并测试了其对红脉穗螟的致病力。通过DNA提取、ITS序列扩增得到的片段长度为555 bp,经比对后发现该菌株与绿僵菌相似度达99%。实验室测定结果表明,处理后14 d,在孢子含量为1×10^(4)个/mL条件下对红脉穗螟幼虫的累积死亡率为62%;在1×10^(8)个/mL时幼虫的校正死亡率可达98.89%,LT50为2.27 d。本研究结果表明,绿僵菌菌株Red-3对红脉穗螟具有一定的致病力,具有应用于红脉穗螟生物防治的潜力。 Tirathaba rufivena Walker(Lepidoptera:Pyralidae)is an important pest of crops such as coconut,areca nut,etc.in Hainan Province,which has caused serious damage and economic losses.In this article,a strain of Metarhizium anisopliae Red-3 was isolated from dead red palm weevil and was identified by morphological characteristics and molecular biological methods.Its pathogenicity to T.rufivena was tested.An approximately 555-bp fragment was amplified from the DNA extract of strain Red-3 by ITS PCR.Further phylogenetic analysis revealed that Red-3 was evolutionarily close to M.anisopliae,with 99%sequence similarity.Laboratory bioassay results showed that,after being treated with the conidia of Red-3 at a concentration of 1×10^(4) conidia/mL for 14 days,the cumulative mortality rate of the T.rufivena larvae was 62%.At a concentration of 1×10^(8) conidia/mL,the Red-3 treatment could result in 99%mortality of T.rufivena larvae with the LT50 of 2.27 days.In summary,the results showed that the M.anisopliae strain Red-3 has certain pathogenicity to T.rufivena and has potential in the biocontrol of T.rufivena.
