新疆某马场马乳头瘤病毒1型全基因组测定及分析 被引量:1

Identification and the Complete Genome Analysis of Equus Caballus Papillomavirus Type 1 in a Farm of Xinjiang

摘要 为探明引起新疆昌吉某马场呼吸困难纯血种公马鼻腔内乳头瘤样组织的病原,采集马鼻腔内乳头瘤样组织样品,采用高通量测序检测样品携带病毒多样性。结果显示,在乳头瘤样组织中检测到马乳头瘤病毒1型,命名为G2株。采用PCR方法扩增该病毒的全基因组序列,结果显示,EcPV-1 G2株与EcPV-1、EcPV-2-9参考株E6、E7、E1、E2、E4、L2和L1基因的核苷酸序列分别为99.0%~99.8%和30%~60%,氨基酸序列同源性分别为97.7%~99.2%和12.1%~56.7%。EcPV-1 G2株和EcPV-1参考株L1基因核苷酸同源性为98.8%~99.1%,表明该毒株为EcPV-1新型变异株。分离获得EcPV-1株并对其全基因组序列进行分析,可为新疆地区EcPV-1流行病学研究提供参考依据。 We applied viral metagenomics to identify potential pathogens,which caused nasal papilloma-like tissues of thoroughbred stallion in a farm in Changji,Xinjiang.The metagenomics data revealed the presence of sequences matching to Equus caballus papillomavirus type 1(EcPV-1)genome sequences(named EcPV-1 G2).After amplification,sequencing and splicing,the complete genome of EcPV-1 was obtained,Sequencing and comparison of the E6,E7,E1,E2,E4,L1 and L2 genes between EcPV-1 G2 and EcPV-1 reference strains,showed that the nucleotide and amino acid identity were 99.0%-99.8%and 97.7%-99.2%,respectively.The E6,E7,E1,E2,E4,L1 and L2 genes of EcPV-1 G2 shared a nucleotide identity of 30%-60%and an amino acid identity of 12.1%-56.7%with EcPV-2-9 reference strains,respec-tively.The nucleotide identity of L1 gene was 99.5%between EcPV-1 G2 and EcPV-1 reference strains,indicating that EcPV-1 G2 was a novel EcPV-1 variant.Our study first identified EcPV-1 and took whole genome analysis,These findings provided fundamental data for the epidemiological study of EcPV-1 in Xin-jiang.
