HPLC同时测定宫炎康胶囊中的8种成分

Simultaneous determination of eight constituents in Gongyankang capsules by HPLC

目的采用HPLC法同时测定宫炎康胶囊中菊苣酸、洋川芎内酯H、洋川芎内酯Ⅰ、洋川芎内酯A、瑟丹酸内酯、6-姜酚、8-姜酚、10-姜酚的含量。方法采用Agilent Zorbax SB-C_(18)色谱柱(250 mm×4.6 mm,5μm),柱温25℃,流动相为乙腈-0.2%冰乙酸,梯度洗脱,流速0.9 mL·min^(-1),检测波长分别为330 nm(菊苣酸)、280 nm(洋川芎内酯H、洋川芎内酯Ⅰ、洋川芎内酯A、瑟丹酸内酯、6-姜酚、8-姜酚、10-姜酚)。结果菊苣酸、洋川芎内酯H、洋川芎内酯Ⅰ、洋川芎内酯A、瑟丹酸内酯、6-姜酚、8-姜酚、10-姜酚的线性范围分别为0.98~19.60、1.56~31.20、1.99~39.80、4.86~97.20、2.27~45.40、7.98~159.60、1.36~27.20、1.74~34.80μg·mL^(-1)(r≥0.9991),平均加样回收率分别为97.77%、98.96%、99.11%、99.84%、98.87%、100.03%、96.85%、96.99%,RSD分别为1.31%、1.09%、0.90%、0.64%、1.24%、0.82%、1.15%、1.11%。结论所用方法操作简便、重复性好,可用于宫炎康胶囊的质量控制。

OBJECTIVE To simultaneous detect chicoric acid,senkyunolide H,senkyunolide I,senkyunolide A,trans-sedanolide,6-gingerol,8-gingerol and 10-gingerol in Gongyankang capsules by HPLC.METHODS The determination was performed on Agilent Zorbax SB-C_(18)(250 mm×4.6 mm,5μm)column with a mobile phase consisting of acetonitrile-0.2%glacial acetic acid solution in gradient elution.The flow rate was 0.9 mL·min^(-1)with the column temperature of 25℃,and the detection wavelength of at 330 nm for chicoric acid,and 280 nm for senkyunolide H,senkyunolide I,senkyunolide A,trans-sedanolide,6-gingerol,8-gingerol and 10-gingerol.RESULTS Chicoric acid,senkyunolide H,senkyunolideⅠ,senkyunolide A,trans-sedanolide,6-gingerol,8-gingerol and 10-gingerol showed good linear relationships within the ranges of 0.98^(-1)9.60,1.56-31.20,1.99-39.80,4.86-97.20,2.27-45.40,7.98^(-1)59.60,1.36-27.20 and 1.74-34.80μg·mL^(-1)(r≥0.9991),respectively.The average recoveries were 97.77%,98.96%,99.11%,99.84%,98.87%,100.03%,96.85%,96.99%with the RSDs of 1.31%,1.09%,0.90%,0.64%,1.24%,0.82%,1.15%,1.11%,respectively.CONCLUSION The method is easy to operate and repeatable,which can be used for the quality control of Gongyankang capsules.
