摘要
目的分离、纯化广西瑶药鸡爪风茎叶中的化学成分,并考察化合物的体外抗肿瘤活性。方法采用硅胶、Sephadex LH-20葡聚糖凝胶柱层析技术对鸡爪风茎叶醇提物进行分离纯化,并利用质谱、核磁共振等波谱数据鉴定化合物结构。通过MTT法测定化合物对肿瘤细胞HCT116和HepG2的抑制作用。结果从鸡爪风中分离得到6个化合物,鉴定了其中5个,分别为:5-羟基-7,8-二甲氧基黄酮(1)、5,6-二羟基-7-甲氧基黄酮(2)、5-羟基-6,7-二甲氧基黄酮(3)、β-谷甾醇(4)、苯甲酸(5)。其中化合物3对HCT116及HepG2细胞作用72 h的IC 50分别为80.67μg·mL^(-1)、45.48μg·mL^(-1)。结论化合物1首次从假鹰爪属植物中分离得到,化合物1、2首次从鸡爪风中分离得到。化合物3对肿瘤细胞HCT116、HepG2具有一定的抑制活性,且对HepG2作用较强。
OBJECTIVE To isolate,purify and identify the chemical constituents from the stems and leaves of the Desmos chinensis Lour.And to investigate the antitumor activity of the compounds in vitro.METHODS The chemical constituents were isolated and purified by silica gel and Sephadex LH-20 and their structures were elucidated with spectroscopic analysis including 1H-NMR,13C-NMR and ESI-MS.MTT method was used to determine the inhibition rate of the compounds on HCT116 and HepG2.RESULTS Six compounds were isolated and five of them were identified as 5-hydroxy-7,8-dimethoxyflavone(1),5,6-dihydroxy-7-methoxyflavone(2),5-hydroxy-6,7-dimethoxyflavone(3),β-sitosterol(4),Benzoic acid(5).The IC50 of the compound 3 was 85.67μg·mL^(-1) and 45.48μg·mL^(-1) for HCT116 and HepG272 h,respectively.CONCLUSION The compound 1 was isolated from this genus for the first time;compounds 1&2 were obtained from Desmos chinensis Lour.for the first time.And the compound 3 exhibited certain cytotoxicity against HCT116 and HepG2 cell lines.
