HPLC法测定大柴胡颗粒中大黄酚含量

Determination of Chrysophanol in DaChaiHu GranuLes by HPLC

目的考察大柴胡颗粒中相关成分的稳定情况,对其中的大黄酚进行测定。方法采用HPLC法对样品进行测定,色谱柱采用C 18柱(250 mm×4.6 mm,5μm),流动相为甲醇-0.1%磷酸(75∶25),检测波长254 nm,流速1.0 mL·min^(-1),柱温28℃,进样量20μL。结果线性范围为大黄酚:0.05588~0.5588μg(R=0.9985),平均回收率为95.96%,RSD=0.59%。结论大柴胡颗粒中大黄酚含量为0.78 mg·g^(-1)。

OBJECTIVE To investigate the stability of related components in DaChaihu granules and to determine Chrysophanol in them.METHODS Samples were analyzed on Fortis C18 column(250 mm×4.6 mm,5μm).Methanol-0.1%phosphoric acid solution(75∶25)served as themobile phase.The wavelength of detection was at 254 nm.The flow rate was 1.0 mL·min^(-1),the column temperature was set at 28℃and the injection volume was 20μL.RESULTS Chrysophanol showed good linearity in the range of 0.05588-0.5588μg(R=0.9985),and the average recovery rate of was 95.96%(RSD=0.59%).CONCLUSION The content of Chrysophanol in DaChaiHu granules was 0.78 mg·g^(-1).
