超滤法测定伏立康唑的结合型药物浓度及血浆蛋白结合率 被引量:1

摘要 目的建立一种测定伏立康唑结合型药物浓度和蛋白结合率的方法。方法采用超滤法处理血浆样品,采用高效液相色谱法结合固相萃取法测定人血浆中伏立康唑的总药物浓度和结合型药物浓度,计算血浆蛋白结合率。结果在总血浆药物浓度为0.1~10.0μg·mL^(-1)范围内,伏立康唑血浆蛋白结合率为62.35%±5.62%,日内、日间精密度RSD<5%。结论本研究所建立的测定方法便捷、灵敏、准确,可用于测定伏立康唑的人血浆蛋白结合率。 OBJECTIVE To establish a method for the determination of bound drug concentration and plasma protein binding rate(PPB)of voriconazole.METHODS An ultrafiltration procedure was applied to pretreat plasma sample,high performance liquid chromatography(HPLC)combined with solid phase extraction method was set up to measure the concentrations of bound and total voriconazole in human plasma,then the PPB was calculated.RESULTS The human PPB of voriconazole was 62.35%±5.62%,in the totol plasma concentration range of 0.1~10.0μg·mL^(-1),the intra-and inter-day precison were less than 5%.CONCLUSION The described method offers a convenient,sensitive and accurate analysis for PPB of voriconazole in human plasma.
