电针对高脂肥胖大鼠脂质蓄积及瘦素抵抗的影响

摘要 目的:观察电针对高脂饮食诱导的肥胖大鼠脂质蓄积对改善瘦素抵抗的影响。方法:选用3周龄SD雄性大鼠42只,随机分为2组:正常组(n=6),普通饲料喂养,高脂组(n=36),高脂饲料喂养。12周后将高脂组造模成功的大鼠(n=18)随机分为模型组、电针组和假针刺组。测定各组大鼠体质量、脂肪质量、血脂、瘦素等相关指标,qPCR检测各组大鼠下丘脑瘦素受体的mRNA表达变化,用HE染色观察比较各组大鼠肝脏的形态学差异。结果:与正常组比较,高脂模型组体质量显著升高(P<0.01),显示肥胖大鼠造模成功。与模型组相比,电针组大鼠的体质量、内脏脂肪量、TG、TCL、leptin均显著降低(P<0.05),下丘脑瘦素受体mRNA表达量显著上升(P<0.01)。HE染色结果显示,电针组大鼠肝脏内的脂滴空泡较模型组明显减少,形态恢复与正常组接近。假针刺组大鼠除项后脂肪量外其他各项指标与模型组相比无明显差异(P>0.05)。结论:电针可以减少肥胖大鼠脂质堆积并改善瘦素抵抗,具有一定的抑制肥胖效果。 Objective:To evaluate the influence of EA on fat accumulation and leptin resistance of diet induced obese rats.Methods:42 SPF SD male rats(3 weeks)were randomly divided into 2 groups:normal diet group(n=6)and high-fat diet group(n=36).The obesity model of high-fat diet group was established by feeding a high fat diet for 12 weeks.Then,the obese rats(n=18)were randomly divided into three groups.model group(n=6),EA group(n=6)and sham-acupoint group(n=6).Then the body weight,fat weight,serum lipid and leptin levels of each group were measured.The leptin receptor mRNA expression in hypothalamus were examined with qPCR.The morphological differences of liver in each group were examined with HE stain.Results:Compared with the normal group,weight gain was significantly increased in the model group(P<0.01),which showed that the obese model was successfully established.Compared with the model group,the body weight,visceral fat weight,TG,TCL and leptin level of EA group were significantly decreased,while the mRNA expression of leptin receptor in hypothalamus significantly increased in EA group.The result of HE stain showed that the lipid vacuoles in the livers collected from EA group were significantly decreased compared with the model group,and the morphology of the liver from EA group was similar to that from the normal group.There was no significant difference between the model group and the sham-acupoint group in terms of related indicators except for brown adipose tissue.Conclusion:EA could reduce obesity by attenuating fat accumulation and improving leptin resistance in obese rats.
