摘要
以双孢蘑菇(Agaricus bisporus)As2796菌株为试材,采用单因素试验和响应面法,研究了摇瓶培养双孢蘑菇液体菌种的最佳条件,以期为高效制备双孢蘑菇液体菌种和还原型液体菌种提供参考依据。结果表明:在加富培养基中,培养料浸提液添加量为50%,过氧化钙浓度为0.010 0 g·(100mL)^(-1),250 mL三角瓶装液量为80 mL时,双孢蘑菇液体菌种的生物量达到0.95 g·(100mL)^(-1),比加富培养基提高了57.3%。得到的液体菌种经过抽滤浓缩后,用胶Y固化制备的还原型液体菌种,4℃保存30 d,仍保持固化前的生长速度。
Taking Agaricus bisporus strain As2796 as the test material,the optimal cultivating conditions for Agaricus bisporus liquid inoculum in shake flasks were investigated by single factor experiment combined with response surface method,in order to provid a reference for the efficient preparation of Agaricus bisporus liquid incoulum and reduced liquid incoulum.The results showed that addition of the compost water extract 50% and calcium peroxide 0.010 0 g·(100 mL)^(-1) in the enriched medium and the liquid volume 80 mL in the 250 mL triangular bottle was the optimal cultivating conditions,the biomass of the mushroom liquid spawn reached 0.95 g·(100 mL)^(-1),which was 57.3% higher than that of enriched medium.The liquid spawn was concentrated by suction filtration and solidificated with glue Y genarating reduced liquid inoculum.After stored at 4°C for30 days,the growth rate of the reduced liquid inoculum was similar with the liquid spawn before solidification.
