摘要
目的检测分析人参、黄连有效成分及其对α-葡萄糖苷酶抑制作用。方法采用HPLC-MS联用法对人参黄连提取物进行成分的分析鉴定,并应用酶底物反应法对其主要成分进行α-葡萄糖苷酶活性抑制试验。结果分别对人参提取物、黄连提取物进行检测分析,共鉴定出11个化合物,包括8个人参皂苷和3个生物碱,进一步选取了含量较高的3种人参皂苷和2种生物碱进行体外α-葡萄糖苷酶的抑制试验,结果表明,人参皂苷和生物碱都是有效的潜在降糖成分,但黄连生物碱具有明显的抑制作用,而人参皂苷Rb1、人参皂苷Rg2和人参皂苷Re对α-葡萄糖苷酶的抑制作用较差。结论本研究初步分析了人参黄连降糖的活性成分,证实了参连饮在实际应用中的可行性和可靠性。
Objective To determine and analyze the active components of ginseng and golden thread and their inhibitory effect onα-glucosidase.Methods The components of ginseng and golden thread extracts were analyzed and identifi ed by HPLC-MS,and the main components were tested for the inhibition ofα-glucosidase activity by the enzyme substrate reaction method.Results A total of 11 compounds were identifi ed from the two kinds of extracts,including 8 ginsenosides and 3 alkaloids,and 3 ginsenosides and 2 alkaloids with higher content were further selected for in vitroα-glucosidase inhibition test.The results showed that ginsenosides and alkaloids were both effective and potential hypoglycemic components,but the alkaloids from golden thread had a more significant inhibitory eff ect,while ginsenoside Rb1,ginsenoside Rg2 and ginsenoside Re had a poorer inhibitory eff ect onα-glucosidase.Conclusion This study has preliminarily analyzed the active ingredients of ginseng and golden thread for reducing blood glucose,and confi rmed the feasibility and reliability of Shenlian Yin in its practical application.
