响应面法优化黑果腺肋花楸花色苷抑菌活性工艺的研究 被引量:8

Optimization of Antibacterial Activity of Anthocyanins from Aronia Melanocarpa by Response Surface Methodology

摘要 以黑果腺肋花楸花色苷(AMA)为原料,研究了AMA对5种食源性致病菌的最小抑菌浓度和最小杀菌浓度;其次以大肠杆菌O157:H7为指示菌,研究了温度、pH值、紫外照射时间、NaCl浓度对AMA抑菌稳定性的影响。试验结果表明,AMA对大肠杆菌的抑菌活性最为明显,其最小抑菌浓度为0.625 mg/mL,最小杀菌浓度为1.25 mg/mL,此外其AMA稳定性会随着温度、pH值、NaCl质量分数的变化而变化,但紫外照射时间对其稳定性基本无影响;应用CCD(中心组合)进行3因素3水平的试验设计,以抑菌圈直径为响应值,进行响应面分析(RSA),结果表明,响应面法优化黑果腺肋花楸花色苷抑菌活性的最佳工艺条件为:温度31.54℃、pH5.26、NaCl浓度1.42%。在此条件下,大肠杆菌经浓度为1×MIC的黑果腺肋花楸花色苷处理后的抑菌圈直径平均可达13.68 mm。 In this study,anthocyanins from Aronia melanocarpa were used as the raw material to study the minimum inhibitory concentration and minimum bactericidal concentration of AMA on five food-borne pathogens.Then E.coli O157:H7 was used as indicator bacteria to study the effects of temperature,pH,UV irradiation time and NaCl concentration on the antibacterial stability of AMA.The results showed that AMA had the most obvious antibacterial activity against E.coli,with the minimum inhibitory concentration of 0.625 mg/mL and the minimum bactericidal concentration of 1.25 mg/mL.In addition,the stability of AMA changed with the change of temperature,pH and NaCl content,but the UV irradiation time had no effect on the stability.The CCD(central combination)was used to carry out the test design of 3 factors and 3 levels,and the response surface analysis was carried out with the diameter of the inhibition zone as the response value(RSA).The results showed that the optimal process conditions for optimizing the antibacterial activity of AMA were as follows:temperature 31.54℃,pH 5.26,NaCl concentration 1.42%.Under these conditions,the average diameter of inhibition zone of E.coli treated with AMA at 1×MIC was 13.68 mm.
