Th17/Treg细胞平衡关系及其相关细胞因子在绝经后骨质疏松症发病机制中的作用研究 被引量:5

摘要 目的:探讨Th17、Treg细胞平衡关系及其相关细胞因子在绝经后骨质疏松发病中可能的作用。方法:随机纳入47例绝经后骨质疏松患者为实验组;且纳入同期49例绝经后健康人为对照组,收集两组研究对象的相关临床资料。流式细胞术(FCM)检测Th17、Treg细胞比例,免疫荧光法(ELISA)检测IL-17、IL-23、IL-6、TGF-β、IL-10的表达,并进行比较和相关性分析。结果:(1)实验组的Th17细胞比例为(2.326±0.378)%,与对照组的(1.421±0.366)%比较均明显增高(P<0.05)。实验组的Treg细胞比例为(3.162±0.816)%,与对照组的(4.469±0.625)%比较均明显降低(P<0.05)。(2)实验组IL-17、IL-23、IL-6表达量与对照组比较均显著升高(P<0.05)。(3)实验组TGF-β、IL-10表达量均低于对照组(P<0.05)。(4)相关性分析显示实验组Th17比例与IL-17呈正相关(r=0.775,P=0.041),与BMD负相关(r=-0.734,P=0.045);实验组患者Treg比例与BMD、TGF-β均呈正相关(r=0.657,P=0.036;r=0.647,P=0.041)。结论:绝经后骨质疏松患者外周血中Th17细胞及其相关的细胞因子增多伴随Treg细胞及其相关分泌细胞因子减少,可能在绝经后骨质疏松骨质免疫损伤中起到一定的作用。 Objective:To investigate the possible relationship between Th17 and Treg cell balance and related cytokines in the pathogenesis of postmenopausal osteoporosis.Methods:47 postmenopausal osteoporosis patients were randomly selected as the experimental group;and 49 postmenopausal healthy persons were included as the control group during the same period.Collect relevant clinical data of two groups of study subjects.Flow cytometry(FCM)was used to detect the proportion of Th17 and Treg cells.Immunofluorescence(ELISA)was used to detect the expression of IL-17,IL-23,IL-6,TGF-βand IL-10.The comparison of measurement data between groups was analyzed by test,and the correlation between variables was analyzed by Spearman correlation.Results:(1)The percentage of Th17 cells in the experimental group(2.326±0.378)%was significantly higher than that in the control group(1.421±0.366)%(P<0.05).The percentage of Treg cells in the experimental group(3.162±0.816)%was significantly lower than that in the control group(4.469±0.625)%(P<0.05).(2)The expression levels of IL-17,IL-23,and IL-6 in the experimental group were significantly higher than those in the control group(P<0.05).(3)The expression levels of TGF-βand IL-10 in the experimental group were lower than those in the control group(P<0.05).(4)Correlation analysis showed that the proportion of Th17 in the experimental group was positively correlated with IL-17(r=0.775,P=0.041),and negatively correlated with BMD(r=-0.734,P=0.045);the proportion of Treg in the experimental group was related to BMD and TGF-βAll were positively correlated(r=0.657,P=0.036;r=0.647,P=0.041).Conclusion:The increase of Th17 cells and related cytokines in peripheral blood of postmenopausal osteoporosis patients,along with the decrease of Treg cells and related secreted cytokines,may play a role in post-menopausal osteoporotic bone injury.
