β-葡聚糖对S180荷瘤小鼠实体瘤抑制作用及其机制研究 被引量:4

摘要 目的研究β-葡聚糖对S180荷瘤小鼠肿瘤抑制作用及其机制。方法将S180细胞接种于小鼠右上肢皮下建立S180荷瘤小鼠模型,随机分为模型组、顺铂组(0.2 mg/kg,腹腔注射)、β-葡聚糖低、中、高剂量(50、100、200 mg/kg)组,每组16只,雌雄对半。隔日给药,连续21 d,ELISA法检测瘤组织、血液及肝脏相关指标表达,Western Blotting检测凋亡蛋白Caspase-3的表达。结果β-葡聚糖三个剂量组均具有明显的抑瘤作用(P<0.01)。与模型组相比,β-葡聚糖三个剂量组脾脏指数均升高(P<0.01);β-葡聚糖三个剂量组AST、ALT、UA和CRE含量下降(P<0.01)。与模型组相比,β-葡聚糖三个剂量组均能增加凋亡蛋白Caspase-3蛋白的表达(P<0.05)。结论β-葡聚糖显著抑制S180腹水瘤荷瘤小鼠的肿瘤生长,其作用机制可能与促进肿瘤细胞的凋亡相关。 Objective To study the inhibitory effect ofβ-glucan on tumor growth in S180 tumor-bearing mice and its mechanism.Methods S180 tumor cells were inoculated subcutaneously in the right upper limb of mice to establish S180 tumor-bearing mice model,which were randomly divided into model group,cisplatin group(0.2 mg/kg,intraperitoneal injection),β-glucan low-,medium-and high-dose(50,100 and 200 mg/kg)groups with 16 mice in each group,intervening at alternative day for 21 days.The tumor tissues,blood and liver were collected to measure the related indexes.Results The 3 doses groups ofβ-glucan had obvious inhibitory effect(P<0.01).Compared with the model group,the spleen index in the 3 doses groups ofβ-glucan increased significantly(P<0.01).Compared with the modelgroup,the contents of AST,ALT,UA and CRE in the 3 doses groups of β-glucan decreased significantly(P<0.01).Compared with the model group,the expression of Caspase-3 protein in the 3 doses groups of β-glucan increased significantly(P<0.05).Conclusionβ-glucan significantly inhibited the tumor growth of S180 tumor-bearing mice,and its mechanism may be related to promoting the apoptosis of tumor cells.
